摘 要:在不均匀牵拉法制动的离体大鼠膈神经膈肌标本上,用微管聚合药红豆生物碱探讨了终板膜的微管在神经肌肉接头乙酰胆碱受体兴奋中的作用。红豆生物碱(20μmol/L,10min)使有神经支配的膈肌终板区乙酰胆碱电位幅度下降了30%;但不影响其时程和膜电位。结果表明该药对神经肌肉接头有突触后抑制作用。其作用机制可能通过影响微管而致使终板膜反应性下降。Taxol was used as a tool reagent and its effect on the function of the micro-tubules ( MTs ) beneath the postsynaptic membrane was studied in an isolated non-uniformly stretched muscle preparation of rat diaphragm. After exposure to taxol (20 μmol/L, 10 min) , the amplitude of acetylcholine potential (AChP) of innervated muscle endplate was decreased by 30% but the time course of AChP and membrane potential remained unchanged. The results indicate that taxol can inhibit the responsiveness of ACh receptor. It is therefore suggested that the site of action of taxol inducing inhibition of postsynaptic responsiveness in neurom-uscular junction may be the MTs beneath the postsynaptic membrane.
