摘 要:目的观察铁离子、乳铁蛋白及其与FeCl3混合溶液对隐形眼镜铜绿假单胞菌生物膜形成的影响。方法通过静态生物被膜形成实验,在铜绿假单胞菌生物膜中加入不同浓度的游离铁、乳铁蛋白及其与FeCl3混合溶液条件下,以倍比稀释平板培养法检测活菌数评价其杀菌活性。再利用扫描电子镜观察正常状态、缺铁环境和部分缺铁环境下铜绿假单胞菌生物膜的微观形态。结果乳铁蛋白对铜绿假单胞菌具有杀菌活性,同时抑制生物被膜形成,其作用与浓度成正相关;FeCl3促进铜绿假单胞菌生长和生物被膜形成;乳铁蛋白与铁结合后其杀菌性减弱,对生物被膜形成的抑制作用减弱。结论铁离子可以促进隐形眼镜铜绿假单胞菌生物膜形成,而一定浓度的乳铁蛋白及其与FeCl3混合溶液形成的缺铁状态,可抑制该生物膜的形成。Objective To study the effects of iron ions,lactoferrin and its mixed solution with FeCl3 on the biomembrane formation of Pseudomonas aeruginosa on contact lenses.Methods Different concentration of free iron,lactoferrin and its mixed solution with FeCl3 were added to biomembrane of Pseudomonas aeruginosa during the static biomembrane formation experimentation.Viable bacterial count was performed using the double dilution method and plate culture assay to evaluate its bactericidal activity.Micromorphology of biomembrane of Pseudomonas aeruginosa on the condition of normal,iron-dificiency and partial iron-dificiency were observed by using scanning electron microscope.Results Lactoferrin showed bactericidal activity toward Pseudomonas aeruginosa and inhibited the biomembrane formation.Its effect had positive correlation with concentration.FeCl3 promoted the growth and biomembrane formation of Pseudomonas aeruginosa.The bactericidal activity and inhibition of biomembrane formation were reduced when lactoferrin combined with iron.Conclusion Iron ions can improve the biomembrane formation of Pseudomonas aeruginosa on contact lenses,which would be inhibited by iron-dificiency resulted from a certain concentration of lactoferrin and its mixed solution with FeCl3.
