摘 要:为研究丙型肝炎病毒血症的诊断,以三组与病毒不同区段互补的引物进行反转录巢式聚合酶链反应检测输血后肝炎17例(急性期14例)。按日本毒株的引物检出35%,按非编码区序列的引物检出76%的HCV RNA,两组引物共检出94%,而按Chiron原型的引物检测6例均阴性。HCV多变异,且血清水平极低,用几组不同保守区段的引物,进行巢式PCR,可以获得较高的检出率。To study the diagnosis of hepatitis C viremia, 17 cases of non-A, non-B post-transfusion hepatitis, including 14 cases during acute phase, were investigated by a reversely transcribed and nested polymerase chain reaction with three sets of primers complemented at different regions of the genome.HCV RNA was detected in 35% of 17 cases with PCR using the primers synthesed according to the sequence of Japanese clone, and in 76% using the primers complemented to the 5' noncoding region of the genome. The total prevalence was up to 94% with thesetwo sets of primers, whereas all were negative in six cases test by PCR with the primers from Chiron prototype sequence. Hepatitis C virus is likely to mutate, and its serum level is quite low, and hence, nested PCR with several sets of primers complemented to the various preserved regions of the genome might be necessary for a higher detection.
