利用SRAP分析岷江下游荔枝资源的遗传多样性与亲缘关系  被引量:9

摘  要:以岷江下游的41份荔枝古树、3份合江荔枝包括合江大红袍、合江糯米糍和楠木叶为试材,选用32对SRAP(Sequence-re-lated Amplified Polymorphism)引物进行PCR扩增。共检测出222条清晰条带,其中产生182条多态性条带,平均每对引物组合5.6条,多态率为81.9%。32对引物组合遗传多样性在0.0329(me18/em26)~0.4990(me21/em18)之间,平均为0.2915,利用NT-SYS-PC(version 2.10s)软件系统进行UPGMA(Unweighted pair group method arithmetic average)聚类分析。结果表明,岷江下游44份材料之间亲缘关系较近,44材料相似系数在0.5586~0.9279,平均相似系数为0.7913。在遗传相似系数0.67处可分成4大类,其中3个引进品种与当地荔枝古树资源明显分开。由此可看出,岷江下游荔枝古树资源是荔枝育种和生物多样性保护的特殊资源。Sequence-related amplified polymorphism(SRAP) markers were employed to detect the genetic variation among 41 samples of ancient litchi trees from the lower reach of Minjiang river and 3 cultivars including Dahongpao,Hejiang nuomici and Nanmuye introduced from Hejiang county in Sihuan province,thirty-two SRAP primer combinations were used,and 182 polymorphic bands out of 222 with an average of 5.6 bands per primer combination were observed.The diversity index of thirty-two SRAP primer combinations varied from 0.0329 to 0.4990 with an average 0.2915,which were analyzed by the unweighted pair group method(UPGMA) analysis.The result showed that 44 accessions had a similarity range from 0.5586 to 0.9279 with a mean of 0.7913.There were four clusters at the similarity level of 0.67,of which 3 cultivars including Dahongpao,Hejiang nuomici and Nanmuye were quite diverse from those ancient litchi trees.It suggested that there were special germplasm resources of ancient litchi in the lower reach of Minjiang river valley for further breeding and biodiversity conservation.
