摘 要:目的建立乳鼠心脏微血管内皮细胞(CMEC)体外培养研究模型。方法采用混合酶(0.06%胰蛋白酶和0.1%Ⅱ型胶原酶均量混合)消化法和差速贴壁法纯化细胞,倒置显微镜对所培养的细胞进行形态学观察,以Ⅷ因子相关抗原免疫组化染色法进行细胞鉴定。结果通过形态学特征及微血管内皮细胞相关特异性抗原检测证实,得到高纯度乳鼠心脏微血管内皮细胞。结论利用混合酶消化法成功分离并培养乳鼠的CMEC,该方法简单易行,可用于进一步的科学研究。Objective To establish the model in vitro of cardiac microvascular endothelial cells (CMEC) in neonatal rats. Methods CMEC were isolated and purified in hybrid enzyme(0.06% trypsin and 0. 1% Ⅱ collagenase) digestion,different centrifugal force and different adherence, then cultured. The cultured cells morphology was observed by inverted microscope. CMEC were identified by immunohistochemical staining for factor Ⅷ related antigen. Results CMEC were confirmed by morphological characteristics and CMEC associated antigen testing. High purity CMEC were obtained. Conclusion CMEC of neonatal rats were successfully isolated and cultured by hybrid enzyme digestion. The method is simple, can be used for further scientific research.
