摘 要:目的 观察低强度红激光照射(Low Power RedLaser Irradiation,LPRLI)后局部血管组织Ⅰ型胶原(Col Ⅰ)mRNA、细胞间黏附分子-1(ICAM-1)的表达和血管平滑肌细胞(VSMC)凋亡,探讨LPRLI抑制动脉粥样硬化兔血管成形术后收缩性血管重塑的可能机制。方法 建立动脉粥样硬化兔的血管损伤模型。100只新西兰白兔随机分为单纯血管拉伤组和单纯拉伤+激光照射组。对术后3天、1周、2周、4周、8周动物组织提取总 RNA、半定量 RT-PCR观察 Col mR-NA表达和免疫组织化学-LSAB法测定ICAM-1、p53、bcl-2/bax。TUNEL法检测VSMC凋亡并计算凋亡指数。结果①Col Ⅰ mRNA的表达:术后3天,两组均可见到Col Ⅰ mRNA的表达,但无统计学意义;术后第 1~4周,Col Ⅰ mRNA的表达LPRLI组明显低于对照组;第8周,两组Col Ⅰ mRNA的表达均减弱,组间无统计学差异。②ICAM-1的表达:免疫组织化学检测术后3天即可看到ICAM-1的表达,LPRLI组明显低于对照组;第1周到第4周,LPRLI组的ICAM-1虽然逐渐增强,但仍然显著低于对照组?Objective In this study, we investi-gated whether LPRLI (Low Power Red Laser Irradiation) could attenuate the expressions of Col I mRNA, ICAM-1, p53, bcl-2/bax and induce apoptosis of VSMC in the rabbit abdominal aorta injury model. Methods We estab-lished atherosclerotic arterial stenosis models successfully.100 male New Zealand white rabbits were randomized into two groups: balloon injury group and balloon injury plus laser irradiation group (total irradiation density: 0. 9J/ cm2). After 3 days, 1 week, 2 weeks, 4 weeks, 8 weeks, abdominal aortas were harvested for reverse tran-scription-polymerase chain reaction (RT-PCR) studies and immunohistocheical studies on the expressions of Col I mRNA, ICAM-1, p53,bcl-2/bax. The presence of apoptotic VSMC was demonstrated by terminal deoxynucleotidyl transferase-mediated dUTP nickl end labeling (TUNEL) method. Results (1)The expression of Col I mRNA at 3rd day after balloon injury, the two groups (LPRLI group vs control group) did not show a significantly difference [(114*24) cmp,vs (118* 11)cmp, P >0.05]. from 1st week to 4th week, the expressions of Col I mRNA showed a significantly difference. The den-sitometric data of RT-PCR in LPRLL group was lower than the control group [(760* 119) cpm vs (879* 121) cpm, P<0.05;(1 120*300) cpm vs (1409*297) cpm, P <0.05; (918 * 180) cpm vs (1109 * 117) cpm, P <0.05];(2)The expression of ICAM-1 from 3rd day to 4th week, the expressions of ICAM- 1 in LPRLI group was significantly reduced both at the intima and ad- ventitia[(0.90*0.48) vs (1.79*0.91), P<0.05; (1.41*0.89) vs (3.46*1.57), P<0.01; (2.60* 0.91) vs (3.51*0.99), P<0.05;(2.31*1.00)vs (2.57 * 1.20), P < 0.05](3)Apoptosis of VSMC. These studies suggested that there were histochemical evidence of apoptosis in all of the injured vessels. There were no quantitative differences in the amount of LPRLI and control vessels at 3 days, 1 week and 8 weeks. But there were marked differences between the two groups. The index of apoptotic VSMC in LPRLI group was higher than control group
