摘  要:目的研究一种纳米金快速增强金标免疫层析实验灵敏度的试验方法。方法以尿HCG金标免疫层析实验为基础,以改性溶液做预处理,盐酸羟胺还原氯金酸制备纳米金颗粒,提高金标免疫层析实验灵敏度。结果盐酸羟胺还原氯金酸制备纳米金颗粒作为增敏试剂,提高胶体金早早孕测试纸检测尿HCG的灵敏度,增敏后HCG检测下限达0.5mIU/ml,比试纸条说明书标注的下限25mIU/ml减小50倍,在15~20min可以完成,实验结果快速、稳定、可靠。结论盐酸羟胺还原氯金酸制备纳米金颗粒可快速提高金标免疫层析试验的灵敏度,结果准确可靠,有望为适时、快速的现场检测方法提供更高灵敏度的技术支持。OBJECTIVE To study a new assay for enhancing the sensitivity of the gold immunochromatographic assay quickly by Au nanoparticles. METHODS The urine human chorionic gonadotropin hormone (HCG) was qualitative detected by gold immunochromatographic assay ( GICA). Based on this, the reaction bands of GICA were treated by modification solution, bound the colloidal gold prepared by NH 2 OH·HCl and HAuCl 4 and the sensitivity was evaluated pre-post enhancing test. RESULTS The sensitizing reagent that prepared by NH 2 OH·HCl and HAuCl 4 was able to be applied to enhance the sensitivity of urine HCG tested by GICA and the detection limit post enhancement was 0.5mIU / ml while the detection limit written on instruction manual of HCG GICA was 25mIU / ml, the sensitivity was increased by 50 times and the assay could be completed in 20-30 minutes also the results was stable and reliable. CONCLUSION That the colloidal gold prepared by NH 2 OH·HCl reducing HAuCl 4 can be applied as sensitizing reagent, it can enhance the sensitivity of GICA quickly and the result was stable and reliable. It may provide a new high sensitive technical support to the poin-of-care testing.
