循经取穴针刺对心肌缺血大鼠模型心肌内向整流钾通道基因表达的影响  被引量:4

摘  要:目的:观察针刺不同经脉经穴对心肌缺血大鼠模型心肌细胞内向整流钾离子通道Kir2.1、Kir2.2mRNA表达水平的影响,从基因水平揭示循经取穴对心肌缺血的作用机制。方法:采用随机数字表法将60只健康雄性SD大鼠随机分成对照组、模型组、内关穴组、列缺穴组、非经非穴组,每组12只。除对照组外,其他组采用皮下多点位注射盐酸异丙肾上腺素(85 mg/kg)建立心肌缺血大鼠模型;对照组在相同部位注射等量生理盐水。造模成功后,给予相应穴位的电针刺激,每天1次,1周后应用实时荧光定量聚合酶链反应技术(Real-timePCR)检测左心室Kir2.1、Kir2.2mRNA的表达水平。结果:与对照组比较,模型组、内关穴组、列缺穴组、非经非穴组Kir2.1、Kir2.2mRNA的表达显著降低(P<0.01)。与模型组比较,内关穴组、列缺穴组Kir2.1、Kir2.2mRNA的表达显著升高(P<0.01)。与内关穴组比较,列缺穴组、非经非穴组Kir2.1、Kir2.2mRNA的表达显著降低(P<0.01)。与列缺穴组相比,非经非穴组Kir2.1、Kir2.2mRNA的表达显著降低(P<0.05)。结论:循经取穴可逆转缺血心肌细胞内向整流钾离子通道Kir2.1、Kir2.2mRNA表达的下降,证实针刺手厥阴心包经的内关穴抗心肌缺血具有循经特异性。Objective:To observe the effect of needling different acupoints on the expressions of Kir2.1, Kir2.2mRNA in myo- cardial cells'transient IKir channel in the rat model of myocardial ischemia, the author designed the research to explore the mecha- nism about needling acupoints along meridians to treat myocardial ischemia on the level of genes. Methods : With random number table ,60 healthy male SD rats were randomly divided into blank group, model group, Neiguan(PC6) point group, Lieque (LU7) point group and neither meridians nor points group ( 12 rats in each group). Except the blank group, the other four groups had sub- cutaneous multi-point injection of Isoproterenol Hydrochloride (85 mg/kg) to establish the rat model of myocardial ischemia;while the blank group were injected with normal saline at the same area. Then we gave the rats electro-acupuncture stimulation at the corresponding acupoints once a day. One week later,we detected the expression levels of Kir2.1 and Kir2.2mRNA of the left ven- tricular applying real-time fluorescence quantitative polymerase chain reaction( Real-timePCR). Results:Compared with the blank group, the expressions of Kir2.1 and Kir2.2mRNA in the model group, Neiguan (PC6) point group, Lieque (LU7) point group, and neither meridians nor points group were significantly reduced ( P 〈 0. 01 or P 〈 0. 05 ). Compared with the model group, the Kir2.1 and Kir2.2mRNA expressions of Neiguan ( PC6 ) point group and Lieque ( LIg7 ) point group were significantly increased ( P 〈 0. 01 or P 〈 0. 05 ). Compared with Neiguan ( PC6 ) point group, the Kir2.1 and Kir2.2mRNA expressions of Lieque ( LU7 ) point group and neither meridians nor points group were significantly reduced ( P 〈 0.01 or P 〈 0. 05 ). Compared with Lieque (LU7) point groups, the Kit2; 1 and Kir2.2mRNA expression of neither meridians nor points group was significantly reduced ( P 〈 0. 01 or P 〈 0. 05 ).
