摘  要:探究了人体胎盘组织AC133+造血干/祖细胞(HSPC)的集落形成能力。选用新鲜的人胎盘组织制成单个细胞(有核细胞)悬液,经流式细胞术(FCM)分析其AC133+造血干/祖细胞的纯度,采用免疫磁性分选法(MACS)分选AC133+和AC133-HSPC,分别进行粒-单集落形成单位(CFU-GM)、红系集落形成单位(CFU-E)、混合集落形成单位(CFU-Mix)培养,以评价其增殖分化能力。结果显示胎盘组织AC133+细胞百分率为4.11%,其细胞亚群集落培养形成CFU-GM的集落数为(37±3)/1×103;形成CFU-Mix的集落数为(34±5)/1×103;形成CFU-E集落数为(33±4)/1×103。胎盘AC133-细胞未见集落形成。结论:人胎盘组织含HSPC,AC133+细胞具有分化为CFU-GM、CFU-Mix、CFU-E的能力;AC133膜抗原可作为胎盘HSPC的分选标志。To study the colony formation capability of AC133+ cells in human placenta tissues. Single cells (nucleated cells) suspension was made from fresh human placenta tissue, the purity of AC133+ hematopoietic stern/progenitor cells were analyzed by the flow cytometry (FCM). The AC133+ and AC133- HSPC cells were sorted by MACS and then cultured by the granulocyte/macrophage colony forming unit (CFU-GM), colonyforming unit erythroid (CFU-E) and mixed forming unit (CFU-Mix) respectively for evaluating their proliferation and differentiation. In human PT, there was 4.11% AC133+ cells, the number of colonies of CFU-GM, CFU-Mix and CFU-E were (37±3)/1*103, (34±5)/1* 103 and (334±) 1 * 103 respectively. No colony ofAC 133- HSPCs was found in the human placenta tissue. There were HSPCs in the human placenta tissue. The PT AC133+ HSPCs cells can differentiate into CFU-GM, CFU-E, CFU- Mix. Thus suggesting that PT AC133+ cells contain HSPCs and the AC133 membrane antigen could be the sign of sorting the HSPCs in PT.
