摘 要:目的:研究不同KIF11蛋白结构域对细胞增殖的影响,并检测对人HEK293T细胞增殖的抑制效果。方法:PCR扩增全长KIF11编码的mRNA,将含有HA抗原表位的KIF11-motor、-tail等结构域克隆到真核表达载体中。观察这些结构域在HEK293T中的表达和对细胞增殖的影响。结果:表达载体转染后,KIF11-motor、-tail能在细胞内有效表达,且都能抑制细胞增殖。结论:KIF11-tail抑制细胞增殖的能力比KIF11-motor更有效。Objective: To investigate the effect of KIFll domains on cell proliferation, and to determine the inhibting effect of HEK293T cells proliferation. Methods: The entire mRNA coding for KIFll was cloned by PCR. HA epitope-tagged KIF11-motor and -tail domains were cloned into an expressing vector. The domains were transfected into HEK293T for examining their ability in inhibiting cell proliferation. Results: The KIFll-motor and -tail domains were successfully expressed and were able to repress cell proliferation. Conclusion: The ability of KIF11-tail domain to inhibit cell proliferation is more than that of KIF 11-motor.
