摘 要:目的评价丹参酮ⅡA后处理对大鼠心肌缺血再灌注损伤的保护作用。方法SD大鼠随机分为假手术组(A组)、缺血再灌注组(B组)和丹参酮ⅡA后处理组(c组),每组8只。A组:制备大鼠心肌缺血再灌注模型,只穿线,不结扎左冠状动脉;B组:缺血再灌注组,缺血40min,再灌注120min;C组:后处理组,结扎左冠状动脉40min,再灌注120min,再灌注前3min和再灌注后2min内静脉注入丹参酮ⅡA。缺血再灌注160min时,经颈总动脉插管抽取动脉血约2ml,离心取血清,测定血清中CK、LDH、MDA和SOD含量。采用TUNEL法观察各组心肌细胞的凋亡指数,并在光镜及电镜下观察细胞形态学变化。结果与B组相比,c组CK和LDH明显降低,凋亡指数亦明显降低;C组SOD活性明显高于B组,MDA含量明显低于B组;c组心肌组织形态学结构得到较大改善。结论丹参酮ⅡA后处理对大鼠心肌缺血再灌注有保护作用。Objective To evaluate protective effects of tanshinone 1I A postconditioning on acute myocardial ischemia/reperfusion injury in isolated rat heart. Methods Twenty-four male SD rats were randomly divided into three groups (shamoperation group, ischemica/reperfusion group and tanshinone II A postconditioning group). Each group has 8 rats. The level of creatikinase (CK), lactate dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde (MDA) were measured. The pathomorphological changes were observed with light microscope and electron microscope. The apoptosis number were determined with the method of TUNEL. Results Compared with group B, the content of CK and LDH were significantly lower in group C. Apoptotic index was significantly reduced. Compared with group B, the activity of SOD was significantly lower in group C, the content of MDA was significantly higher. The differences were all statistically significant. Histomorphology of myocardium under light microscope and electron microscope in group C was improved more distinctly than those in group B after reperfusion for 120 min. Conclusion Tanshinone lI A postconditioning provide obviously protective effect against myocardial isehemia/reperfusion injury in rats.
