摘  要:目的:研究多巴胺D1受体的选择性激动剂SKF 38393对大鼠伏隔核棘状神经元自发兴奋性突触后电流的影响。方法:大鼠伏隔核切片,全细胞膜片钳记录神经元兴奋性突触后电流。结果:SKF 38393在较低浓度(100 nmol/L)时,增加自发兴奋性突触后电流频率,不改变其幅度;在较高浓度时(10μmol/L)时,对自发兴奋性突触后电流频率无明显影响,而显著升高其幅度。结论:D1受体激动剂对大鼠伏隔核棘状神经元谷氨酸受体介导的突触传递起双向的调节作用。Objective: To study the effect of dopamine D1 receptor selective agonist SKF 38393 on spontaneous excitatory postsynaptic current(sEPSC) in medium spiny neurons of rat's nucleus accumbens. Methods: Rat nucleus accumbens slices were prepared. The EPSCs were investigated using whole-cell patch-clamp recording in medium spiny neurons. Results: At lower concentration ( 100 nmol/L) , SKF 38393 increased the frequency of sEPSCs, without changing its amplitude. At higher concentration ( 10 μmol/L), SKF 38393 did not induce obvious change in frequency of sEPSCs, but increased the amplitude obviously. Conclusion : D 1 agonist may have bidirectional regulating actions on glutamate receptor-mediated synaptic transmission to medium spiny neurons of rat's nucleus accumbens.
