摘 要:目的:观察黄连对铜绿假单胞菌(Pseudomonas aeruginosa,PA)生物学特性(毒力因子、生物被膜、菌体运动)的影响,探讨黄连对铜绿假单胞菌的影响及机制。方法:铜绿假单胞菌与不同浓度的黄连共同培养,刚果红降解试验检测弹性蛋白表达活性;氯仿盐酸法测定绿脓菌素分泌量;结晶紫染色法观察生物被膜起始量;利用黄连琼脂平板检测菌体集群运动(swarming)与泳动(swimming)。结果:黄连在12.5mg/ml和6.25mg/ml浓度下对绿脓菌素分泌有显著抑制,生物被膜在25mg/ml至6.25mg/ml时受到显著抑制;各组弹性蛋白酶表达均水平较正常对照组显著降低。细菌在12.5mg/ml与6.25mg/ml条件下泳动能力与集群运动均明显弱于正常对照。结论:黄连对铜绿假单胞菌多种生物学特性产生抑制作用,能够对受密度感应系统高度调控的弹性蛋白酶产生显著抑制,提示黄连抑菌机制包括对抑制密度感应系统产生抑制效应,黄连可能是铜绿假单胞菌密度感应抑制剂。To investigate the effects of Chinese goldthread against Pseudomonas aeruginosa virulence, motility and biofilm formation, regulated by quorum sensing system. Method: Expression of elastase and pyocyanin were assessed quantitatively by using elastin Congo red and ultraviolet spectrophotometry at different concentrations of goldthread. The biofilm formation was detected by Crystal violet staining under the same cultivation condition. Swarm and swim motility assays were undertaken in petfi dishes. Result: Expression of elastase and pyocya- nin were decreased respectively at concentration of both 12.5mg/ml and 6. 25mg/ml while the biofilm formation exhibited the same trend ofinhibition. The motility assays were reduced at both concentrations of 6.25mg/ml and 12.5mg/ml. Conclusion: Goldthread could inhibit the virulence expressions, the biofilm formation and the swimming motility, mainly regulated by QS system. The combination of methods could be applied preliminarily in screening of quorum sensing inhibitors.
