摘 要:目的:探讨巴豆霜、大黄及其成分是否通过维持CD4+CD25+Treg平衡,发挥溃疡性结肠炎(UC)大鼠的治疗作用。方法:采用2,4,6-三硝基苯磺酸(TNBS)法复制UC大鼠模型,将40只大鼠随机分为空白对照组、模型组、巴豆霜+大黄组、巴豆霜+没食子酸组、巴豆霜+大黄素组。观察结肠大体形态损伤、组织学变化,测定各组大鼠外周血CD4+CD25+Treg、CD4+CD25+Foxp3+Treg淋巴细胞变化。结果:与正常组比较,模型组大鼠外周血Treg细胞降低显著(P<0.01)。与模型组相比,巴豆霜+大黄组、巴豆霜+没食子酸组结肠大体形态损伤、组织学变化显著改善。治疗1周后,和模型组比较,巴豆霜+大黄组,巴豆霜+没食子酸组大鼠外周血Treg细胞有显著增高(P<0.01或P<0.05),且两组间比较没有差异;而巴豆霜+大黄素组没有差异(P>0.05)。结论:巴豆霜+大黄、巴豆霜+没食子酸对大鼠UC具有显著治疗作用,提高大鼠外周血CD4+CD25+Treg、CD4+CD25+Foxp3+Treg细胞可能是其作用机制之一,没食子酸是大黄和巴豆霜配伍发挥止泻作用的有效物质之一。To explore the therapeutic effect of Semen Crotonis Pulveratum, Rhubarb and it's component by keeping CIM + CD25/CIM + Treg, CIM + CIY25 + Foxp3/CD4 + Treg balance in Wister rats with ulcerative colitis (UC). Methods: Ratsi were randomized into 5 groups: normal control group, the model group, Semen Crotonis Pulveratum + Rhubarb,Semen Crotonis Pulveratum + Emodin, Semen Crotonis Pul- veratum + Gallic acidgroup with 10 rats in each group and UC was induced by administrating 2, 4, 6tfinitrobenze-sulphonic acid (TNBS) in rats. Rats were sacrificed to observe the general morphology and histopathological changes of the colon, and detected the percentages of CD4 + CD25 + Tceg,CIM+ CIY25 + Foxp3 + Treg in peripheral blood. Results: Compare to the model group, abnormally macroscopical, histological changes were improved in BDS + DH,and BDS + MSZ groupso The percentages of CIM + CD25 + Foxp3/CIM+ Treg in peripheral blood in the model group is significantly lower than those in normal control group ( P 〈 0.01 ) ,while significantly high After treating one week in BDS + DH, BDS + MSZ groups ( P 〈 0.01 or P 〈 0.05 ). No improvement or change was found in group BDS + DHS. Conclusion: BDS + DH, BDS + MSZ have positive effects in treating colitis in rats by increasing the percentages of CD4 + CD25 + Foxp3/CIM + Treg in peripheral blood.
