抑郁模型大鼠海马CA3区突触结构的体视学变化及加味温胆汤的作用

摘　　要：目的:探讨抑郁模型大鼠海马CA3区突触结构的体视学变化及加味温胆汤的作用。方法:以孤养结合慢性不可预见性应激抑郁模型大鼠为研究对象,在实验第7d,14d,21d,28d,检测各组大鼠海马突触结构的体视学变化。结果:在抑郁形成过程中,21d时,与空白组比较,模型组大鼠突触活性区长度、突触后致密物质厚度明显减少,突触间隙宽度增加明显。28d时,差异进一步明显。随着加味温胆汤(12g/kg)治疗周次的增加,抑郁大鼠突触活性区长度、突触后致密物质厚度减少,以及突触间隙宽度上升的趋势得到抑制,与模型组比较,在21d时,有明显统计学差异(P<0.05),28d时,差异增大(P<0.01)。加味温胆汤汤组(12g/kg)与氟西汀组(1.8mg/kg)相比无明显统计学意义。结论:抑郁症可引起海马神经元突触结构损伤,导致出现神经元萎缩,加味温胆汤对此有逆转作用。To probe stereological changes of hippocampal synaptic structure in CA3 region of rat depression model and results after reg- ulating by Jiawci Wcndan decoction. Methods： The isolated model control group was living under unpredictable chronic stress. At the 7th, 14th, 21st, 28th day of the experiment, we detected stereological changes of hippocampal synaptic structure of each group. Resulis： During the progress of depression, there were some tendencies： At the 21 st day, comparing with the blank group, the length of active synaptic region in model control group was shorter, the thickness of post-synaptic dense area started to thin out gradually and the synaptic cleft was widen; At the 28th day, there were increasing differences between the model control group and the blank group. With the therapic weeks prolonging by Jiawei Wendan decoction（ 12g/kg）, these tendencies appeared on depression rats can be controlledl Compared to model control group, there existed significant differences at the 21st day（ p 〈 0.05 ） ;and these significant differences became larger at the 28th day（ P 〈 0.01 ）. Conclu- sion： stereological damages of hippecampal synaptic structure resulting from depression led to neuron atrophy, which could be reversed by Jiawei Wendan decoction.
