参术胶囊对胃癌细胞MKN45显微结构和超微结构的影响

摘　　要：目的:研究参术胶囊对胃癌细胞MKN45显微结构和超微结构的影响及参术胶囊能否诱导MKN45细胞凋亡。方法:用姬姆萨染色技术研究参术胶囊对MKN45细胞株的一般形态学产生的影响;用电子显微镜技术研究参术胶囊对MKN45细胞株超微结构的影响。结果:光镜下姬姆萨染色后观察显示4.4、22、44、220、440μg/ml参术胶囊作用24h后,部分细胞变小,着色较深、结构混乱、染色质边聚。电镜显示4.4、22、44、220、440μg/ml参术胶囊能造成细胞核内染色质聚成团块,核膜完整,细胞内线粒体肿胀,胞浆内空泡增加。结论:参术胶囊能抑制胃癌细胞株MKN45生长,其机制与诱导细胞凋亡有关。可能是通过线粒体途径诱导细胞凋亡的。To study effects of Shenzhu Capsules on the mierostructure and ultrastructure of gastric cancer cell line MKN45. Methods： The Giemsa＇s staining was used to study the general effect of Sbenzhu Capsules on MKN45 cell line; the electron microscope technique was ap- plied to study the effect of Sbenzhu Capsules on the microstructure of MKN45. Results：The result of MTT showed after 24h action of Sbenzhu Capsules 4.4 、22 、44 、220,440μg/ml, the OD values of MKN45 was lower; the Giemsa＇s staining showed that after 24h action of Sbenzhu Capsules 4.4、22、44、220,440μg/ml, some cells turned smaller, darker, confused in the structure and had chromosome-margination. The result of the electron microscope technique showed clustered chromosomes, integrity membrane , swelling mitochondria and increased intraey- toplasmic vacuoles in MKN45. Conclusions： Shcnzhu Capsules could inhibit the growth of the gastric cancer cell line. The mechanism was related to cell apoptosis. The mitochondrial pathway might be the right one Shenzhu Capsules induced apoptosis by.
