摘  要:目的:探讨温补肾阳方对心力衰竭大鼠心肌能力代谢的影响及可能的作用机制。方法:采用冠状动脉前降支结扎法制备心力衰竭大鼠模型,造模成功后随机分为肾阳高、中、低剂量组及假手术组、模型组和曲美他嗪组,术后2周开始灌胃,共给药8周。8周之后采用高效液相法测定心肌组织中的ATP、AMP和ADP含量,并采用分光光度计测量心衰大鼠心肌中Ca2+-Mg2+ATP酶、Na+-K+ATP酶和SDH含量。结果:温补肾阳方17.4 g/kg,8.7 g/kg,4.4 g/kg对大鼠心肌中ATP含量较模型组显著升高,AMP含量较模型组显著降低,Ca2+-Mg2+ATP酶、Na+-K+ATP酶和SDH含量明显高于模型组;温补肾阳(17.4 g/kg,8.7 g/kg)大鼠心肌中ADP含量较模型组降低。结论:温补肾阳方对心力衰竭大鼠心肌的能量代谢有一定改善作用。To investigate the effect and its mechanism of Warming Kidney on myocardial energy metabolism of heart failure rats. Meth- ods : The heart failure rats was established by ligating left anterial descending coronary artery. Make sure the success of model. Rats were di- vided into high, middle and low doses of Warming Kidney group , sham operation normal control group , model group and trimetazidinetria- zine control group randomly. Rats were gavaged with Warming Kidney 2 weeks later and continued 8 weeks. After 8 weeks, we detect the content of ATP, AMP and ADP in myocardial tissue by HPLC. Record the level of Ca2+ -Mg2+ ATPase, Na+ -K+ ATPase and Succinate De- hydrogenase through the spectrophotometer. Results: The content of ATP in the myocardium of each Wanning Kidney group( 17.4 g/kg,8. 7 g/kg,4.4 g/kg), compared with the model rates, significantly increased( P 〈 0.01 ). And the content of AMP significantly decreased( P 〈 0. 01 ). The Na + -K + ATPase, Ca2. -Mg2 + ATPase and Succinate Dehydrogenase activity in each group of Wanning Kidney were significantly higher than the model group (P 〈 0.01 ). The content of ADP in Warming Kidney high and middle groups( 17.4 g/kg, 8.7 g/kg)was lower than model group ( P 〈 0.05 ). Conclusion: Wanning kidney can improve the myocardial energy metabolism of rats with heart failure.
