摘  要:目的:观察三七总皂苷对肝纤维化大鼠的保护作用及其可能的作用机制。方法:大鼠随机分成正常组、模型组、三七总皂苷(50、100、200 mg/kg)组和秋水仙碱(Col,0.1 mg/kg)组。除正常组外,其余各组采用皮下注射50%四氯化碳(CCl4)每周2次连续18周的方法诱导肝纤维化大鼠模型。于造模第9周起,给药组分别灌胃给予相应的受试药物,正常组和模型组给予等量溶媒,每天1次,连续10周。实验结束后,放射免疫法测定血清透明质酸(HA)、层粘连蛋白(LN)、III型前胶原氨端肽(PIIINP)、Ⅳ型胶原(CIV)的含量;比色法测定肝匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)的水平;同时取固定部位肝脏组织,HE及Masson染色观察肝纤维化程度;免疫组织化学法检测肝组织Bcl-2、Bax蛋白的表达。结果:与模型组相比,三七总皂苷(100、200 mg/kg)不仅可减轻大鼠肝纤维化程度,降低HA、LN、PIIINP、CIV和MDA含量,升高SOD水平,还可抑制肝纤维化大鼠肝组织中Bax的表达,促进Bcl-2的表达。结论:三七总皂苷具有一定的抗大鼠肝纤维化作用,上调Bcl-2的表达,下调Bax的表达,抑制肝细胞的凋亡可能是其作用机制之一。Objective:To observe the effect of Panax notoginseng saponins(PNS) on experimental liver fibrosis in rats and to explore its possible mechanisms.Methods: The rats were randomly divided into six groups,including normal group,liver fibrosis model group,PNS(50,100,200 mg/kg) treated groups and Col(0.1 mg/kg) group.Rat liver fibrosis was induced by 50% CCl4 twice a week for 18 weeks.PNS were used daily via lavage at 9th week for 10 weeks.The hyaluronic acid(HA),laminin(LN),procollagen III N-terminal peptide(PIIINP),collagenⅣ(CIV) level in serum were determinate by radioimmunity,malondialdehyde(MDA) and(superoxide dismutase) SOD were determinate by chromatometry.Moreover,liver samples were taken to examine the degree of liver fibrosis by HE and Masson staining and stained by immuninochemistry of Bcl-2,Bax.Results: As compared with the fibrotic model group,PNS(100,200 mg/kg) significantly reduced histopathological change,degraded HA,LN,PIIINP,CIV and MDA contents,steped up the level of SOD.Meanwhile PNS could effectively prevent the expression of Bax,enhanced the expression Bcl-2 in liver fibrosis rats.Conclusion: PNS can resist hepatic fibrosis possibly by up-regulating Bcl-2 expression and down-regulating Bax expression to inhibit liver cell apoptosis.
