摘 要:目的:探讨山药多糖抗神经细胞缺氧性凋亡作用。方法:研究分为正常对照组、凋亡诱导组、山药多糖0.025g/L组、山药多糖0.05g/L组、山药多糖0.1g/L组、山药多糖0.25g/L组。采用"Neurobasal+B27"体外神经细胞无血清培养,免疫细胞化学鉴定神经细胞,MTT测定药物毒性,AnnexinV/PI双染流式细胞仪及Hochest33342荧光染色检测细胞凋亡。结果:山药多糖在2.0g/L之内预处理神经细胞对其无毒性作用,山药多糖在0.05g/L~0.1g/L范围显著地改善缺氧对神经细胞生长的抑制,0.025g/L~0.25g/L范围内显著地抑制缺氧复氧诱导的神经细胞早期凋亡。结论:山药多糖在一定浓度范围内具有抗缺氧复氧诱导的细胞早期凋亡作用。Objective:To explore the effect of Chinese Yam polysaccharide on preventing the neuron apoptosis indued by hypoxia.Methods: The study was divided into normal control group,apoptosis positive group induced by hypoxia,experiment 1 group treated by 0.025g/L Chinese Yam polysaccharide,experiment 2 group treated by 0.05g/L Chinese Yam polysaccharide group,experiment 3 group treated by 0.1g/L Chinese Yam polysaccharide group,experiment 4 group treated by 0.25 g/L Chinese Yam polysaccharide group.Neural cells were cultured by Neurobasal + B27 in serum-free in vitro.Neural cells were identified by immunocytochemistry.Drug toxicity was determined by MTT assay,Neuron apoptosis was examined by Annexin V-FITC/PI in flow cytometry and Hoechst 33342 fluorescent staining.Results: The percentage of neural cells was 93.70% ± 0.56% in serum-free culture.The Chinese Yam polysaccharides was non-toxic for neural cells pretreated within 2.0 g/L.The Chinese Yam polysaccharides significantly prevented the inhibition of neural cell growth from hypoxia from 0.05g/L to 0.1g/L,(P 0.05).Early apoptosis of neural cells induced by hypoxia-reoxygenation was significantly inhibited by Chinese Yam polysaccharides from 0.025g/L to 0.25g/L(P 0.05).Conclusion: The Chinese Yam polysaccharide has an effect on inhibitting early apoptosis of neural cells induced by hypoxia-reoxygenation in a certain concentration range.
