摘 要:目的:研究瓜蒌皮对急性心肌缺血心肌损伤的作用效果,初步探讨其机制。方法:结扎大鼠冠脉动脉,制备冠心病急性心肌梗死模型。丹参片制成水悬液,作为阳性对照药,瓜蒌皮制成水煎液,大鼠灌胃给药。氯化三苯四氮唑染色,测定心肌梗死率;生理记录仪记录心电图变化,检测心脏功能;生化试剂盒检测血浆中肌酸激酶、肌酸激酶同工酶、谷草转氨酶、羟基丁酸脱氢酶和乳酸脱氢酶5项心肌酶活性,确定心肌细胞坏死程度;生化试剂盒检测心肌组织中丙二醛含量及超氧化物歧化酶活性,明确大鼠心肌清除氧自由基能力。结果:对急性心肌缺血大鼠,瓜蒌皮水煎液起效剂量为30g/kg,可显著降低其心肌梗死率,维护缺血心肌正常生理功能,明显降低血液中5项心肌酶活性,增强心肌组织超氧化物歧化酶活性,降低丙二醛含量,作用效果与丹参片无显著性差异。结论:瓜蒌皮水煎液对大鼠急性心肌缺血有明显保护作用,其机制可能与增强清除自由基能力有关。Objective:Traditional Chinese Medicine Trichosanthes pericarpium results from the pericarp of Trichosanthes kirilowii Maxim or Trichosanthesost hornii Harms.This herbal medicine had been used to cure coronary heart disease patients for two thousand years in China.This study aims at to evaluate the pharmacodynamical effect in myocardial infarction protection and to research into the underlying mechanism of this protection.Methods: To make acute myocardial ischemia animal model,1 hour after the last administration,the left anterior descending coronary branch were ligated in male Wistar rats that have received intragastrical administration for 4 days.Rats in different groups were given Trichosanthes pericarpium decoction,the Chinese nostrum Fufangdanshenpian(the positive control drug),and 0.9 % sodium chloride solution.6h after of operation,myocardial tissue was stained with triphenyltetrazolium chloride(TTC).The degree of myocardial infarction was evaluated by electrocardiograph(ECG),the infarction area and the infarction rate.To evaluate the myocardial injury,serum levels of creatine-kinase(CK),creatine-kinase-MB(CK-MB),lactate dehydrogenase(LDH) and aspartate-amino transferase(AST) were detected in these different groups of rats.Moreover,in order to find out the mechnism by which Trichosanthes pericarpium to protect myocardial infarction,the content of malondialdehyde(MDA) and superoxide dismutase(SOD) in myocardial tissue were measured.Results: The effective dose of the Trichosanthes group is 30g/kg.Compared with that in the model group rats,the myocardial infarction rate of rats in the Trichosanthes group decreased evidently(P 0.05),the serum activity of CK,CK-MB,LDH,AST and α-HBDH were also obviously low(P 0.05 or P 0.01).The myocardial MDA content was markedly reduced,but the activity of SOD in the myocardial increased observably(P 0.05).Conclusion: The protective effect of Trichosanthes pericarpium on the rats acute myocardial ischemic is significant,t
