摘 要:目的∶探讨川贝母止嗽颗粒对急性支气管模型大鼠的影响。方法:制备大鼠急性支气管炎模型并灌胃川贝母止嗽颗粒18.6g生药/kg、9.3g生药/kg、4.65g生药/kg,采用病理组织学方法观察大鼠气管、支气管的病理改变,透射电镜观察大鼠的支气管超微结构改变,Elisa法测定大鼠血清IL-8、TNF-α、NF-kB,sICAM-1的含量。结果∶川贝母止嗽颗粒18.6g生药/kg、9.3g生药/kg、4.65g生药/kg大鼠支气管光镜下偶见或见少量炎细胞浸润,大鼠积分光密度值、目标面积显著降低;川贝母止嗽颗粒18.6g生药/kg、9.3g生药/kg支气管纤毛上皮线粒体正常,4.65g生药/kg支气管纤毛上皮线粒体轻度肿胀;18.6g生药/kg、9.3g生药/kg均能显著降低大鼠血清中IL-8、TNF-α、NF-kB、sICAM-1的含量,4.65g生药/kg能显著降低大鼠血清中TNF-α、NF-kB、sICAM-1的含量。结论∶川贝母止嗽颗粒18.6g生药/kg、9.3g生药/kg、4.65g生药/kg均能明显改善急性支气管炎模型大鼠的支气管病理状态,改善支气管的超微结构,下调大鼠血清中升高的IL-8、TNF-α、NF-kB,sICAM-1。Objective: To study the effects of Bulbus Fritillariae Cirrhosae cough relieving granule on rats of acute bronchitis model. Methods: Animal model of acute bronchitis rats was established and administered with Bulbus Fritillariae Cirrhosae cough relieving granule at doses of 18. 6,9. 3,4. 65g crude drug/kg respectively. The pathlogy change of rats tracheobronchial was observed by the method of histopathology; the ultrastructure changes of bronchi of rats was oberved by transmission electron microscopic,using Elisa method for determination of IL-8,TNFα,NF-kB,sICAM-1 in serum of rats. Results: Less or small amount of inflammatory cell infiltration of rats bronchus could be seen under light microscope and the IOD and target area of inflammatory cell infiltration significantly decreased in the doses of 18. 6,9. 3,4. 65g crude drug/kg; mitochondrial of bronchi ciliated epithelium shows normal status in the doses of 18. 6,9. 3g crude drug/kg but mild swelling in the dose of 4. 65g crude drug/kg; the level of IL-8,TNF-α,NF-kB,sICAM-1 could be significantly decreased in the doses of 18. 6,9. 3,4. 65g crude drug/kg,the level of TNF-α,NF-kB,sICAM-1 could be significantly decreased in the dose of 4. 65g crude drug/kg. Conclusion: The pathological state and ultrastructure changes bronchi of rats could be reduced and the level of IL-8,TNF-α,NF-kB,sICAM-1 could be downregulated by Bulbus Fritillariae Cirrhosae cough relieving granule.
