退银汤对大鼠的长期毒性研究

摘　　要：目的:退银汤连续灌胃给药90天对大鼠的长期毒性,评价其安全性。方法:采用100、200、400g生药/kg 3个剂量组给大鼠连续灌胃3个月,正常对照组给予等容量的生理盐水。给药结束后,随机取2/3大鼠(雌雄各半)进行血液细胞学、血液生化学、体质量、脏器质量、脏器系数和常规检查及病理组织学检查,剩余大鼠停药观察1个月,再检测上述各项指标。结果:各剂量组大鼠外观体征、行为活动、血液细胞学、血液生化学、体质量、脏器质量和脏器系数等均未见异常;心、肝、脾、肺、肾、卵巢、睾丸等脏器组织形态学检查未见与用药有关的病理学改变;可逆性观察表明,退银汤对大鼠未引起延迟毒性。上述各项指标与对照组比较,差异均无统计学意义。结论:退银汤较大剂量和较长疗程服用无明显毒性反应,为临床应用提供了实验依据。Objective：To observe the chronic toxicity of Tuiyin Soup intragastric administrated on rats long-termly,and to evaluate its safety.Methods： The rats were divided into 3 dose groups as 100g,200g and 400g crude drug /kg,and then give the Tuiyin Soup to the rats orally for 3 months.The control group was given the same dose of physiological saline samely.After the last administration,randomly selected 2 /3 rats（half male and half female） to take on blood cytology,biochemistry,body weight,organ weight,organ coefficients,normal test and histopathological examination,Continue to observe the remaining rats while stop using the soup for 1 months,and then detect the indexes.Results： The appearance,behavior,blood cell,blood biochemistry,body weight,organ weight and organ coefficient of each dose group rats showed no abnormality;And there were no significant pathological changes in the heart,liver,spleen,lung,kidney,ovary of the rats related to the soup.The reversible observe showed that the Tuiyin Soup did not cause delayed toxicity to the rats.Compare the indexes of the control group to the test group,the difference was not statistically significant（P 0.05）.Conclusion： There is no obvious toxic reaction in the use of high dose and long course of treatment with Tuiyin Soup,provides the experimental evidence for clinical application.
