摘 要:目的:探讨加味苓桂术甘汤对代谢综合征模型大鼠糖代谢紊乱的调节作用。方法:大鼠随机分为正常对照组,高脂高盐(模型)组,二甲双胍组,阿托伐他汀组,加味苓桂术甘汤大、中、小剂量组。除正常对照组外,其他各组大鼠均以高脂高盐饲料喂养。并于模型制作当日各给药组大鼠分别ig给予药物治疗,每日1次,连续8w。其中降脂组给予阿托伐他汀8mg/kg,降糖组给予二甲双胍670 mg/kg,大、中、小剂量组给予加味苓桂术甘汤浓缩液18g/kg、12g/kg、6g/kg;正常对照组、模型组给予同样量的生理盐水。每10d称量各组动物体重一次;给药最后一天眼球取血测定空腹血糖;采用酶联免疫吸附分析方法测定血清瘦素、胰岛素水平,计算胰岛素抵抗指数。结果:加味苓桂术甘汤能降低代谢综合征模型大鼠体重,能调节血糖水平,其瘦素含量与模型组比较,均显示有非常显著性的差异。能调节胰岛素水平。结论:加味苓桂术甘汤对代谢综合征所致体重增加有降低作用,对由此所致血糖、瘦素、胰岛素含量变化能调节到正常水平,提示加味苓桂术甘汤对代谢综合征所致糖代谢紊乱有调节糖代谢,治疗糖代谢障碍性疾病的作用。Objective:To explore the regulatory effects of reformatted ling guizhugan decoction on glycometabolism disorder in metabolic syndrome.Methods: 70 male specified-pathogens free(SPF) Wistar rats,aged 3weeks,were divided into seven groups in randomized manner: the control group with distilled water diet;the model group with high fat(49% lard) and high salt(4% NaCl);the lipid-lowering group treated with atovastatin;the glycemic-lowering group treated with metformin;the large dose,middle dose and small dose groups treated with reformatted lingguizhu gan decoction.The rats of every group were taken in high fat and high salt except control group.And the rats of every group were taken drugs administration by gavage at the same time at once every day for 8 weeks.Among them,the rats in lipid-lowering group were taken 8mg/kg atovastatin;the rats in glycemic-lowering group were taken 670mg/kg metformin;the rats in reformatted lingguizhu gan decoction large dose group,middle dose group and small dose group were taken 18 g/kg,12 g/kg and 6 g/kg reformatted lingguizhu gan decoction concentrated solution separately.The rats in control group and model group were taken normal salt administration by gavage for equal content.The rats' body weight in every group were weighed;the blood glucose were determinated;the level of serum leptin and insulin were detected by enzyme-linked immunosorbent assay(ELISA).And insulin resistance index(HOMA-IR) was counted.Results: The body weight changed into slow increase;the blood glucose,leptin and insulin content could be regulated to a level of very significant difference(all P 0.01) while these groups were compared with the model group after reformatted lingguizhu gan decoction were taken for 8 weeks.Conclusion: The reformatted lingguizhugan decoction can lower body weight and blood glucose,regulate leptin and insulin to a normal level in metabolic syndrome model rats while they were changed with glycometabolism disorder.It was suggested that reformatted linggu
