二十五味珊瑚丸对戊四氮致痫大鼠行为学和脑电的影响

摘　　要：目的:观察二十五味珊瑚丸对致痫大鼠行为学和脑电的影响,评价二十五味珊瑚丸的抗痫效果。方法:将65只大鼠随机分5组,即模型组、珊瑚丸低、中和高剂量组(100mg/kg、200mg/kg、400mg/kg)和丙戊酸钠组,模型组大鼠在造模前灌胃给予去离子水,其他各组分别灌胃给药,连续干预7天,末次灌胃1h后,各组大鼠腹腔注射戊四氮,观察其行为学改变,描记脑电图变化。结果:珊瑚丸中、高剂量组及丙戊酸钠组大鼠发作性阵挛潜伏期、痫性发作级别、发作时间、阵挛次数和发作后恢复探索时间明显优于模型组。模型组大鼠阵发出现大量高幅尖波、棘波、棘慢复合波及尖慢综合波,珊瑚丸中、高剂量组和丙戊酸钠组只见极少量的棘波或尖波。结论:二十五味珊瑚丸可改善大鼠急性痫性发作行为的各项指标,抑制脑电痫样放电。Objective：In order to investigate the effect of Coral 25 on the acute seizure model,electroencephalogram of the model were observed after treatment.Methods： 65 of wistar rats were divided into 5 groups,the group of modle（A）,the group of Coral 25 low dose（B）,the group of Coral 25 middle dose（C）,the group of Coral 25 high dose（D）,and the group of valproate acid sodium（E）,and there were 13 rats in each group.All of rats in each group were administered by intragastric administration 7 days at 8： 00 am and 20： 00pm.The group of A was given equal volume of deionized water,while the E was given valproic acid sodium by 200mg /（kg.d）,and the B,C,D are given Coral 25 pill by different concentration,such as 100mg /（kg.d） and 200mg /（kg.d） and 400mg /（kg.d）.After 1 hour of the above administrations,all groups of rats were injected PTZ intraperitoneally,according to 50mg/kg.And then observed the ethology change of the rats immediately in 30 mins and the change of EEG.Results： 1.The incubation period of PTZ acute convulsion could increase in the group of B,C,D and E compared with the group of A（F = 4.698,P = 0.001）,but the group of B,C,D and E has no statistical difference.The three groups of Coral 25 did not had dose-effect relationship.2.The EEG in group of A1（normal group） was base wave,such as α,β wave,and scatered θ wave,and had no obvious rhythm.The EEG in group of A（model group） had a large number of high amplitude sharp waves,spikes,spikeslow wave.The epileptic discharges in group of C,D were inhibited,and there was only a very small number of spikes or sharp waves.The EEG in group of B had more sharp waves spikes,spike-slow wave,but the amplitude was lower than the model group（A）.The EEG in group of E had scattered spikes or sharp waves and low amplitude.Conclusion ： The three groups of Coral 25 can significantly restrain the acute epileptic seizure and brain electrical abnormal discharge of rats.
