摘  要:目的观察徐长卿丹皮酚对兔膝骨性关节炎关节软骨MMP-1和TIMP-1表达水平的影响。方法健康成年雄性大耳白兔40只随机分为正常组、模型组、生理盐水组、曲安奈德组、徐长卿丹皮酚组,每组8只。生理盐水组、曲安奈德组、徐长卿丹皮酚组于模型建立后的第1周分别开始膝关节内注射0.9%生理盐水、曲安奈德、徐长卿丹皮酚各0.2mL/只,每周2次,共10次。用药结束1周后,取右侧股骨内侧髁关节面软骨标本,行HE染色观察形态学改变,RNA原位杂交对兔关节软骨MMP-1和TIMP-1 mRNA作定位和定量分析。结果徐长卿丹皮酚组软骨组织MMP-1mRNA表达水平较正常组增高,较曲安奈德组和模型组低;徐长卿丹皮酚组软骨组织TIMP-1 mRNA表达水平较曲安奈德组增高;曲安奈德组TIMP-1 mRNA表达水平与模型组、生理盐水组组间比较无统计学差异。结论徐长卿丹皮酚能通过抑制损伤软骨中MMP-1的表达水平达到治疗骨性关节炎的作用,有很好的临床推广前景。Objectve To investigate the influence of Cynanehi paniculati paeonol on the expression level of rabbit knee articular cartilage of osteoarthritis MMP - 1 and TIMP - 1. Methods 40 healthy adult male rabbits were randomly divided into normal group, control group, saline group, triamcinolone aeetonide group, Radix Cynanehi paniculati paeonol group, with 8 rats in each group. The physiological saline group, Maria Nade group, Radix Cynanehi panieulati paeonol groups respectively in first weeks after model establishment to the knee joint in- jection of 0.9% saline, Maria Nade, paniculati paeonol each 0.2ml, 2 times / week, a total of 10 times. 1 weeks after the end of treatment, the right medial femoral condylar articular cartilage specimens, immunohis to chemistry staining and observed the morphological changes, RNA in situ hybridization for the localization and quantification of rabbit articular cartilage of MMP - 1 and TIMP - 1 mRNA. Results Ex- pression of Cynanchum paniculatum paeonol group cartilage tissue MMP - 1 mRNA levels higher than the normal control group, compared with triamcinolone aeetonide group low. Expression of cartilage panieulatum paeonol group TIMP - 1 mRNA levels were triameinolone aeetonide group. Conclusion Cynanehi paniculati paeonol can achieve the therapeutic effect on OA by inhibiting expression levels of MMP - 1 damage of cartilage, has good prospects for clinical promotion.
