摘 要:目的:观察健脾化湿法及含药血清对小鼠胰星状细胞活化增殖的影响以筛选出最佳药物效应浓度。方法:体外传代培养小鼠胰星状细胞,采用MTT比色法观察健脾化湿方药血清、卡莫司他药物血清及空白血清对小鼠胰星状细胞活化增殖影响。结果:健脾化湿方药组均能够有效抑制小鼠胰星状细胞的活化增殖,终浓度为10%的健脾化湿方药组与同等浓度的空白血清组比较有差异(P<0.05),与无血清组比较亦同样有统计学意义(P<0.05)。结论:健脾化湿法及含药血清可有效抑制小鼠胰星状细胞的活化增殖,10%是其最佳药物效应浓度。Objective: To investigate the effect of the method of nourishing spleen and resolving dampness on the proliferation of the pancreatic stellate cells,and ascertain the best medical concentration of drug serum.Methods: The mouse pancreatic stellate cells was cultured in vitro.The proliferation of the cells treated with the drug serum containing the agent of nourishing spleen and resolving dampness,the drug serum containing camostat and serum-free were observed.The degree of proliferation was assayed by MTT method.Results: The groups of the method of nourishing spleen and resolving dampness can effectively suppress the activation and proliferation of pancreatic stellate cells.Final concentration of 10% of the group of the method of nourishing spleen and resolving dampness compared with the serum-free group and the same concentration of blank serum group shows difference.Conclusion: The drug serum containing the agent of nourishing spleen and resolving dampness can effectively regulate activation of pancreatic stellate cells proliferation.the drug serum concentration of 10% is the best concentration for drug serum containing the agent of nourishing spleen and resolving dampness regulating the proliferation of the activated mouse pancreatic stellate cells.
