摘  要:目的:观察疏肝健脾和胃安神法对D-IBS大鼠结肠粘膜CRH及CRH-R1 mRNA表达的影响。方法:将11窝育龄7 d的新生SD大鼠110只随机分出10只正常组,余100只利用幼鼠母子分离加直肠刺激法复制大鼠IBS内脏高敏感性模型,造模成功后按体重随机分出模型组、安神和胃法高、中、低剂量组,和西药马来酸曲美布汀对照组各20只。模型复制成功后分别给予蒸馏水、生理盐水、安神和胃法1.52、3.04、6.08 g/kg、舒丽启能0.1 g/kg灌胃,1次/d,连续2 w,治疗后再次评价其腹部收缩反射(AWR)阈值,并运用RT-PCR法检测大鼠结肠粘膜CRH及CRH-R1 mRNA的表达。结果:安神和胃法能够升高内脏高敏感性大鼠AWR阈值,升高结肠粘膜CRH-R1 mRNA的表达。结论:安神和胃法对内脏高敏感型大鼠有较好的治疗作用,其作用机制可能与影响其结肠粘膜CRH-R1 mRNA表达相关。Objective: To explore the effects of the method of mitigating the stomach and tranquilization on the expression of CRH and CRH-R1 mRNA of colon mucosa in rats with D-IBS.Methods: 110 7-postnatal-day neonatal SD rats were randomly divided into the normal group(10 rats) and hypersensitive IBS rats model(100 rats) which was established by the isolation and administration of irritating the rectum,randomly and equally divided the model into the groups of model,high dose,medium dose,low dose and Trimebutine Maleate as the control.The three groups of different dose were treated at the dose of 1.52,3.04 and 6.08 g / kg one time per day while the other three groups were treated with distilled water,saline and Trimebutine Maleate tablet 0.1 g / kg in the same way.Recorded the abdominal withdrawal reflex(AWR) threshold strain and detect the expression of CRH and CRH-R1 mRNA of colon mucosa by the method of PT-PCR after the treatment for continuously 2 weeks.Results: The method of mitigating the stomach and tranquilization could improve the AWR threshold strain and strengthen the expression of CRH and CRH-R1 mRNA of colon mucosa.Conclusion: The method of mitigating the stomach and tranquilization have markedly curative effects to the rats with visceral hypersensitivity,whose function may be related with its affection on the expression of CRH and CRH-R1 mRNA of colon mucosa.
