摘 要:目的:采用中心复合设计—效应面法优化叠鞘石斛中石斛酚的提取工艺。方法:以甲醇体积分数、料液比和超声提取时间为自变量,以石斛酚提取率为因变量,通过对因变量进行多元线性回归拟合,采用效应面法优化最佳提取工艺。结果:确定石斛酚提取工艺为甲醇体积分数50%~61%,料液比1∶19~1∶22,超声提取时间16~21min,工艺验证预测值与实测值的偏差为-1.82%。结论:该方法可用于优化叠鞘石斛石斛酚提取工艺。Objective: To optimize the process of extracting effective constituents of gigantol from Dendrobium denneanum Kerr.by central composite design-response surface.Methods: Methanol concentration,ratio of solvent to raw material and time for ultrasound extraction were selected as independent variables.The overall desirability of the extraction rates of gigantol were set as dependent variables.Linear mathematic models were used to estimate the relationship between independent and dependent variables response surface methodology was used to optimize the process of extraction.Results: The optimal conditions of extraction process were determined as follows: The materia medica was ultrasound extracted for 16 ~ 21 min with 19 ~ 22 fold solvent of 50% ~ 61% mbethanol.The deviation etween observed and predicted values was-1.82%.Conclusion: The central composite design-response surface methodology is convenient and highly predictive for optimizing the extraction process of gigantol.
