螃蟹甲水提取物对溃疡性结肠炎小鼠的治疗作用

摘　　要：【目的】探讨螃蟹甲水提取物对溃疡性结肠炎模型小鼠的作用及其机制。【方法】雄性昆明小鼠给予自由饮用5%的葡聚糖硫酸钠(dextran sodium sulfate,DSS)溶液7d,以制备溃疡性结肠炎模型,同时给予柳氮磺吡啶肠溶片、螃蟹甲水提取物(低、高剂量),实验结束后处死小鼠,取结肠组织,进行疾病活动指数(disease activity index,DAI)及组织学损伤的评估,用ELISA法检测结肠组织匀浆中白细胞介素10(Interleukin-10,IL-10)的含量,用免疫组化法评价IL-10在结肠组织中的原位表达。【结果】螃蟹甲水提取物灌胃使溃疡性结肠炎小鼠的DAI明显降低,肠道组织的病理学明显改善,剂量关系明显;同时使结肠组织中IL-10的含量升高(P<0.05),高剂量与柳氮磺吡啶肠溶片起相当的作用。【结论】螃蟹甲水提取物对DSS诱导的溃疡性结肠炎小鼠有较好的治疗作用,而这种作用可能是通过上调节肠道组织中IL-10的含量来达到的。[Objective] To investigate the effects of Phlomis younghusbandii water extract on ulcerative colitis （UC） in mice and its mechanism. [Methods]The UC mouse modal were induced by adding 5% DDS into the drinking water. After the administration of Phlomis younghusbandii water extract, mice were killed, disease activity index （DAI） were assessed, and pathological sections were used to detect the changes of local intestinal mucosa, ELISA was used to detect the level of IL-10 secreted of intestinal mucosa, and the expression of IL-10 were measured by immunohistochemistry. [ Results] The Phlomis younghusbandii water extract can reduce DAI scores, relief the clinical symptoms, increase the level and expression of IL-10 in colonic tissue （P 〈 0.05）. [Conclusions] Phlomis younghusbandii water extract is much helpful to the UC mouse induced by DDS. The therapeutic effects maybe act through promoting the expression of IL-10.
