补肾健脾方含药血清对人肝癌细胞株SMMC-7721细胞增殖和细胞凋亡的影响

摘　　要：目的:探讨中药补肾健脾方含药血清对人肝癌细胞株SMMC-7721细胞增殖和细胞凋亡的影响。方法:制备补肾健脾方含药血清,分为高、中、低浓度组;另设复方斑蝥组和甲状腺素组作为阳性对照组,并以空白细胞组作为空白对照组。培养人肝癌细胞株SMMC-7721进行体外实验,用四甲基偶氮唑蓝(MTT)比色法检测各组培养24、48、72 h时细胞增殖抑制率,流式细胞仪检测细胞凋亡情况。结果:各药物组含药血清对人肝癌细胞株SMMC-7721的抑制作用与浓度呈负相关、与作用时间呈正相关,在5%浓度、48 h作用下达到抑制率的高峰。中药干预组细胞凋亡率呈现浓度依赖性,高浓度组显著高于低浓度组(P<0.05);复方斑蝥组的细胞凋亡率略低于中药高浓度组(P>0.05),甲状腺素片组略高于中药低浓度组(P>0.05)。结论:补肾健脾方含药血清可抑制人肝癌细胞株SMMC-7721细胞的增殖,具有诱导人肝癌细胞株SMMC-7721细胞凋亡的趋势。Objective： To discuss the effect of ＂Bushen Jianpi Decoction＂ drug serum on proliferation and apoptosis of human hepatoma cell line SMMC-7721.Methods： Human hepatoma cell line SMMC-7721 was cultivated and divided into three groups at high,medium,low concentration.＂Fufang Banmao＂ group and thyroxine group are positive control group,and blank cell group as the blank control group.The SMMC-7721 was cultured in vitro,and the inhibition ratio of proliferation was detected by MTT at 24h,48h and 72h.The apoptosis was detected by FCM.Results： Each drug group containing serum was negatively correlated with inhibition of Human Hepatoma Cell Line SMMC-7721 and the concentration was positively correlated with the duration of action.In the forty-eight hours and in 5% concentration it has the best results.The rate of apoptosis was in a concentration-dependent manner,and the high concentration TCM group was significantly higher than the low one（P＆lt;0.05）;the rate of apoptosis in ＂Fufang Banmao＂ group was slightly lower than that of the high concentration TCM group（P＆gt;0.05）,and Thyroxine group was slightly higher than the low concentration TCM group（P＆gt;0.05）.Conclusion： ＂Bushen Jianpi Decoction＂ can inhibit cell proliferation and has the tendency of inducing apoptosis.
