摘 要:目的:初步探索小柴胡汤诱导SMMC-7721人肝癌细胞的分化机制。方法:通过检测培养的肿瘤细胞在小柴胡汤中药血清处理后光镜形态学变化、胞内LDH和SDH活性以及细胞周期改变研究小柴胡汤对SMMC-7721细胞的诱导分化作用。结果:小柴胡汤中药血清作用于肿瘤细胞后,光镜形态学观察发现细胞有向正常肝细胞分化的趋势;中药血清组肿瘤细胞的LDH活性下降、SDH活性升高,且小剂量血清组作用显著;与生理盐水血清组和空白组比较,中药血清组肿瘤细胞处于G0/G1期的细胞比例明显增加,S期细胞比例下降。结论:小柴胡汤可分别从细胞糖代谢方式和细胞周期调控方面诱导SMMC-7721肿瘤细胞分化。Objective: Preliminary exploring differentiation mechanism of Xiaochaihu Decoction inducing SMMC-7721 human hepatocellular carcinoma cells. Methods: Detecting the morphological changes under optical microscope, Intracellular activity of LDH and SDH, cell cycle changes of cultured tumor cells after treated by Xiaochaihu Decoction drug serum to study differentiation effect. Results: It could be observed differentiation trend that tumor cells turning to normal liver cell by microscopy after treated by Xiaochaihu Decoction drug serum. Intracellular activity of LDH decreasing and Intracellular activity of SDH increasing in the tumor cells of Chinese medicine serum groups, while it could be observed remarkable effect in small dose of Chinese medi- cine serum group. Compared with physiological saline groups and contrast group, the proportion of tumor cells in the G0/G1 phase was significantly increasing, while declining in S phase in Chinese medicine serum groups. Conclusion: Xiaochaihu Decoction can induce SMMC-7721 tumor cells differentiation respectively from the intracellular glucose metabolism and cell cycle regulation.
