摘 要:目的探讨骨髓基质干细胞(BMSCs)与施万细胞(SCs)联合移植对周围神经损伤的修复作用。方法雌性SD大鼠48只随机分为A、B、C、D 4组,每组各12只;制作右下肢长10 mm坐骨神经缺损模型后,A组注入全贴壁法培养的BMSCs与差速贴壁结合阿糖胞苷法培养的SCs,B组仅注入BMSCs,C组仅注入SCs,D组注入磷酸盐缓冲液(PBS)作为阴性对照。分别于4、8周观察动物一般状态,测定坐骨神经功能指数(SFI)、神经传导速度(NCV)以及免疫组化检测表皮生长因子(EGF)、碱性成纤维细胞生长因子(bFGF)的表达水平。结果 4周时,SFI绝对值A组(53.07±5.36)优于B组(62.73±7.63)、C组(67.17±8.06)、D组(77.56±2.79),差异有统计学意义(P<0.05);A组损伤侧NCV[(20.38±3.13)m/s]优于B组[(16.54±2.18)m/s]、C组[(17.74±0.93)m/s]、D组[(8.25±1.73)m/s],差异有统计学意义(P<0.05)。8周时,SFI值A组(35.43±4.5)优于B组(48.25±3.62)、C组(51.48±3.93)、D组(70.53±5.48),差异有统计学意义(P<0.05);A组损伤侧NCV值[(25.67±2.18)m/s]优于B组[(19.69±4.07)m/s]、C组[(21.37±3.17)m/s]、D组[(10.93±1.59)m/s],差异有统计学意义(P<0.05)。4、8周时A、B、C组EGF和bFGF表达平均光密度值(AOD)与D组比较,差异均有统计学意义(P<0.05)。结论 BMSCs与SCs联合移植能够促进损伤轴突的再生,恢复神经功能,对周围神经损伤具有明显的修复作用。Objective To investigate the recovering effect of co-transplanting of BMSCs and Schwann cells for periph- eral nerve injury. Methods 48 female SD rats were divided into group A, B, C and D randomly with 12 rats in each group. The models of 10 mm sciatic nerve defect were manufactured in right lower extremities, BMSCs were cultured by whole bone marrow adherent and SCs were cultured by differential attachment with Ara-C. BMSCs and SCs were injected to group A, BMSCs were injected to group B, SCs were injected to group C, PBS were injected to group D as the negative control, the general states, sciatic nerve functional index (SFI) and nerve conduction velocity (NCV) were measured at the 4th and 8th weeks, and expression levels of bFGF and EGF were detected by immunohistochemistry. Results At the 4th week, the SFI of group A (53.07±5.36) was better than that of group B (62.73±7.63), group C (67.17±8.06) and group D (77.56±2.79), the differences were statistically significant (P 〈 0.05); the NCV of group A [(20.38±3.13)m/s] was superior than that of group B [(16.54_±2.18)rn/s], group C [(17.74±0.93)m/s] and group D [(8.25± 1.73)m/s], and the differences were statistically significant (P 〈 0.05). At the 8th week, the SFI of group A (35.43±4.5) was better than that of group B (48.25_±3.62), group C (51.48-±3.93) and group D (70.53-±5.48), and the differences were statistically significant (P 〈 0.05); the NCV of group A [(25.67±2.18)rn/s] was superior than that of group B [(19.69±4.07)m/s], group C [(21.37±3.17)m/s] and group ]9 [(10.93±1.59)m/s], and the differences were statistically sig- nificant (P 〈 0.05). A OD value of EGF and bFGF expression at the 4th and 8 th week in group A, group B, group C were found had statistically significant differences with group D (/9 〈 0.05).
