摘 要:目的观察瘦素在外周对骨关节炎模型大鼠关节软骨中转移生长因子β表达的影响。方法将60只7周龄雄性Sprague-Dawley大鼠随机分为对照组、骨关节炎对照组(模型组)、骨关节炎实验组(观察组),每组20只,观察组大鼠膝关节中注射100μg瘦素后,取胫骨平台,用反转录-聚合酶链式反应法及免疫组织化学法检测胫骨平台关节软骨中瘦素及转移生长因子β(TGF-β)的变化。结果观察组大鼠胫骨平台关节软骨瘦素、TGF-βmRNA水平与模型组比较差异无统计学意义(P>0.05),模型组、观察组大鼠胫骨平台关节软骨瘦素、TGF-βmRNA水平均较对照组增高(P<0.05)。模型组和观察组大鼠胫骨平台关节软骨瘦素、TGF-β免疫组织化学灰度均低于对照组,差异有统计学意义(P<0.05),而模型组与观察组大鼠比较差异无统计学意义(P>0.05)。结论瘦素在外周对关节具有保护作用,在骨关节炎的病理生理过程中起重要作用。Objective To determine the influence of leptin on the expression of articular cartilage transforming growth factor beta(TGF-β)in rats with osteoarthritis undergoing peripheral function. Methods Sixty 7-week-old Sprague-Dawley rats were divided randomly into three groups, with twenty animals in each group, the control group, the control osteoarthritic group (model group) and the experimental osteoarthritic group( observation group). In observation group, 100μg leptin was injected into the knee joints of rats. Then the tibial plateaus were collected and the changes of leptin and transforming growth factor (TGF-β) in articular cartilage of tibial plateaus were detected by reverse transcriptase-polymerase chain reaction and immuno- histochemical analysis. Results Compared with model group, the mRNA levels of leptin and TGF-β in articular cartilage of tibial plateau in observation group had not significant difference( P 〉 0.05 ). The mRNA levels of leptin and TGF-~ in articu- lar cartilage of tibia] plateau in model group and observation group were higher ( P 〈 0.05 ) , the immune histochemical gray scales of leptin and TGF-~ in articular cartilage of tibial plateau were significantly lower than those in control group ( P 〈 0. 05 ) ;the differences between the model group and the observation group had not statistical significance (P 〉 0.05 ). Conclu- sion The peripheral protective effect of leptin plays an important role in the pathophysiology of osteoarthritis.
