摘  要:目的制作一种稳定的中度弥漫性轴索损伤(DAI)动物模型,为深入研究DAI提供有用工具。方法健康成年雄性Sprague-Dawley大鼠75只,完全随机分为A、B、C 3组,每组25只,参照Marmarou等方法改造致伤装置,分别采用70 cm×400 g、90 cm×400 g和110 cm×400 g的冲击力打击大鼠头部,制成DAI模型。观察大鼠伤后生命体征及神经系统表现、病死率、脑组织标本形态及病理学特征。结果 3组受伤大鼠的生命体征及神经体征均有不同程度的改变,组织形态学证实均存在弥漫性脑损伤特征,与冲击力呈正相关。与B组和C组比较,A组病理损害程度较轻,不具有广泛的DAI典型病理特征。A、B、C 3组的病死率分别为0%、8%、28%,B组与C组之间的病死率比较差异有统计学意义(P<0.05)。结论采用90 cm×400 g冲击力可建立稳定的大鼠中型DAI模型,病理学特征明显,且稳定性好。Objective To make a stable animal model of moderate diffuse axonal injury(DAI) , and provide useful tool for further studying DAI. Methods A total of 75 healthy male adult Sprague-Dawley rats were randomly divided into group A, B, and C, with 25 rats in each group. The animal models were established by beating the rats' head with the modified Marmarou's traumatic device,the impact forces were 70 cm ~400 g in group A,90 cm x400 g in group B,and 110 cm ~400 g in group C. The vital signs, nervous system signs,mortality, and the characteristic of morphology and pathology of brain tissue samples of all the rats were observed. Results The different degrees' changes in vital signs and nervous system signs had been observed in all injured rats. Diffuse brain injury was confirmed by histomorphology, whose severity degree positively related to impact force. Pathological lesion in group A was slighter than that in group B and group C, and there was no wide typical patho- logical features of DAI in group A. The mortalities of group A,B,and C were 0% ,8% ,and 28% ,respectively. The mortality had statistical significance between group B and group C ( P 〈 0.05 ). Conclusion A stable rat model of moderate DAI can be established with impact force of 90 cm× 400 g, its pathological features are typical, and its stability is good.
