Effects of Electroacupuncture at Different Points on Mesenteric Microcirculation and the VIP Content of Different Viscera in COPD Rats

摘  要:目的观察电针丰隆(ST40)、尺泽穴(LU5)对慢性阻塞性肺病(chronic obstructive pulmonary diseases,COPD)大鼠肠微循环及不同脏器血管活性肠肽(vasoactive intestinal peptide,VIP)分布的影响,观察同经穴与相关经穴对肺及肠的影响特点,探讨穴位相对特异性的规律。方法将32只wistar雄性大鼠随机分为正常组、模型组、尺泽组和丰隆组,每组8只。除正常组外,其余各组均采用气管滴入LPS配合烟熏方法制备COPD模型。丰隆组和尺泽组隔日针刺1次,共治疗12次,实验周期为28 d。观察各组大鼠肠系膜微血管管径和血液流态的变化,并采用放射免疫法测定肺、肠及下丘脑中VIP含量。结果与正常组相比,模型组大鼠肠系膜微血管血流显著加快(P<0.05),丰隆组显著减慢(P<0.01):与模型组相比,尺泽组及丰隆组大鼠血流均显著减慢(P<0.01)。各组大鼠肠、下丘脑中VIP含量与同组肺中VIP含量比较,差异均具有统计学意义(P<0.01,P<0.05)。尺泽组大鼠下丘脑中VIP含量与模型组比较,差异具有统计学意义(P<0.05)。结论尺泽、丰隆穴均可阻逆COPD引起的肠系膜微血管血流加快。尺泽穴可显著降低下丘脑中VIP含量,提示尺泽穴阻逆COPD引起的血流加快与尺泽穴降低下丘脑中VIP含量有关。Objective To observe the effects of electroacupuncture at points Fenglong(ST40) and Chize(LU5) on mesenteric microcirculation and the distribution of vasoactive intestinal peptide (VIP) in different viscera in rats with chronic obstructive pulmonary diseases (COPD), investigate the characteristics of the influences of homo-meridian points and related meridian points on the lung and intestine and explore regularities in the relative specificities of acupuncture points. Methods Thirty-two male Wistar rats were randomly allocated to normal, model, Chize and Fenglong groups, 8 rats each. A COPD model was made by intratracheal instillation of LPS with the cooperation of fume smoking in all groups except the normal group. Fenglong and Chize groups were given acupuncture once every other day, for a total of 12 treatments. The experimental period was 18 days. Mesenteric microvascular caliber and blood flow were measured in each group of rats. Pulmonary, intestinal and hypothalamic VIP contents were determined by radioimmunoassay. Results Compared with the normal group, blood flow velocity increased significantly in the model group (P〈0.05) and decreased significantly in the Fenglong group (P〈0.01). Compared with the model group, blood flow velocity decreased significantly in the Chize and Fenglong groups (P〈0.01). There was a statistically significant difference between intestinal or hypothalamic VIP content and pulmonary VIP content in each group of rats (P〈0.01,P〈0.05). There was a statistically significant difference in hypothalamic VIP content between the Chize and model groups of rats (P〈0.05). Conclusions Points Chize(LU5) and Fenglong(ST40) can both prevent a COPD-caused increase in mesenteric microvascular blood flow velocity. Point Chize(LU5) can significantly reduce hypothalamic VIP content, suggesting that point Chize(LU5) preventing a COPD-caused increase in blood flow velocity is related to it reducing hypothalamic VIP content.
