摘  要:目的对升麻葛根汤经方颗粒汤剂、配方颗粒汤剂与传统汤剂七种化学成分含量进行测定、比较。方法采用高效液相色谱法,Elite SinoChrom ODS-AP色谱柱(250 mm×4.6 mm,5μm),色谱系统Ⅰ:以乙腈-(B)0.02%醋酸水溶液为流动相,梯度洗脱,检测波长230 nm(葛根素、芍药内酯苷、大豆苷、芍药苷、甘草苷);色谱系统Ⅱ:以乙腈-水溶液为流动相,梯度洗脱,检测波长245 nm(升麻素、大豆苷元)。结果 7种成分的线性关系良好,精密度、重复性、稳定性的RSD都低于3%,加样回收率为98.3%~103.6%;传统汤剂与经方颗粒汤剂含量较为接近,而配方颗粒汤剂中的含量要高于与传统汤剂和配方颗粒汤剂。结论该文建立的含量测定方法简便、快捷、准确,为升麻葛根汤的进一步研究提供了依据。【Objective】To compare the seven chemical components in traditional decoctions,the single-flavor granules and formula granules of Shenma Gegen decoctions.【Methods】 The samples were separated on Elite SinoChrom ODS-AP column(250 mm×4.6 mm,5μm).Chromatographic system Ⅰ: the mobile phase consisted of acetonitrile and 0.02% acetic acid using a gradient elution,the UV wavelengths switch had been detected by 230 nm(puerarin,albiflorin,daidzen,paeoniflorin,liquiritin);Chromatographic systemⅡ: The mobile phase consisted of acetonitrile and water using a gradient elution,the cimifugin and aidzein had been detected by 245 nm.【Results】 All the 7 essential compounds showed good linearity in the rang of the testing concentration.The RSD of the precision,reproducibility and stability tests were less than 3%,the recovery was 98.3%~103.6%.Compared with the ingle-flavor granules,the content of the traditional decoction and the formula granules was lower.【Conclusion】The method is simple,rapid and accurate for comparison of the seven constituents in the three different forms,it can provide the scientific basis for further research of the shen ma ge gen decoction.
