摘  要:目的建立红松松塔多糖含量测定方法。方法采用改良差示酚硫法测定红松松塔多糖含量。结果多糖溶液沸水浴30min水解完全,显色液预配,用量为5.0mL;显色后的待测液在2h内稳定;测定波长λmax=490nm;线性范围为3.57~21.42μg·mL-1,r=0.999;回收率为98.3%,RSD为1.06%。结论该方法可用于红松松塔多糖的含量测定,含杂质的样品不必进行繁琐的预分离纯化,方法简便、准确,重复性好。Objective To establish a method to determine the contents of polysaccharides from pine cone of Pinus koraiensis or from preparations. Methods The polysaccharides from pine cone were determined by an.improved-differential phenol sulfuric method. Result Polysaccharides were hydrolyzed to monosaccharide in boiling water bath for 30 min. 5.0 mL of the color liquid was used. The color was stable within 2 h;the calibration curve was linear in the range from 3.57 to 21.42μg·mL^-1 ,with r 0. 999. The average recovery was 98.3% with RSD 1.06%. Conclusions The irnproved-diferential phenol sulfuric method is simple, precision and repeatable for the determination of polysaccharides from pine cone of Pinus koraiensis and from preparations of pharmaceutics that contain impurities.
