摘  要:目的提取分离香椿树皮中有降血糖活性作用的总黄酮。方法将香椿树皮总黄酮粗提物(含生药材2g·mL-1)上D101大孔吸附树脂分离纯化,以树脂径高比、上样流速、洗脱流速、洗脱剂用量为主要影响因素,采用L9(34)正交实验优化最佳分离工艺。结果最佳分离工艺参数为:树脂径高比为1∶12,上样流速为10mL.min-1,体积分数70%乙醇的洗脱流速为1.0L.h-1,洗脱剂用量为2BV。结论 D101大孔吸附树脂对香椿树皮总黄酮的分离效果较好,适用于工业化生产。Objective To study the separation process of total flavonoids from the bark of Toona sinensis. A. Juss. , which have the effect of decreasing blood glucose. Methods The bark total flavonoids crude extract (containing raw medicinal herbs 2 g · mL^-1 ) was separated by D101 macroporous resin. The resin column,the sample flow rate, the elution flow rate and the elution volume were used as the main influencing factors in L9 (34 ) orthogonal experiments to optimize the optimal separation process. Results The optimal separation parameters were as follows: diameter and length ratio of resin column 1 : 12,the sample flow rate 10 mL ·min^-1 , the volume fraction 70% ethanol, the elution flow rate of 1.0 L · h^-1 ,and elution solvent ammount 2 BV. Conclusion The D101 macroporous resin is suitable for the industrial production applications.
