国产依替巴肽抗犬血小板聚集作用研究

摘　　要：目的研究国产血小板糖蛋白Ⅱb/Ⅲa受体拮抗剂依替巴肽体内、体外抗犬血小板聚集作用及作用特点。方法采用比浊法测定血小板聚集,观察依替巴肽体外抗血小板聚集作用;模拟临床采取负荷量与维持量连续给药的方式,观察依替巴肽体内抗血小板聚集作用。结果依替巴肽体外抗犬血小板聚集的半数抑制浓度为136.7±72.6 nmol·L-1;体内给药可迅速抑制犬血小板聚集,在给药期间维持其抑制效应并呈现剂量依赖性;各剂量组血小板聚集抑制率均大于80%,达到临床血小板抑制靶目标。结论国产依替巴肽体内、体外可强效、快速、稳定地抑制犬血小板聚集。Objective To observe the inhibitory effects of national made eptifibatide on platelet aggregation in canine. Method Turbi- dimetie method was used to measure the platelet aggregation to assess the antiplatelet efficacy of national made eptifibatide in vitro and ex vivo. Results Our data suggested that national made eptifibatide inhibited ADP-induced canine platelet aggregation in vitro with ICs0 value of 136.7 ±72.6 nmol · L^-1. It also significantly inhibited ADP-induced platelet aggregation in canine dose-depend- ently ex vivo. The inhibitory rate on platelet aggregation reached 80 %. Conclusion National made eptifibatide shows a fast, marked and sustained inhibitory effect on platelet aggregation in canine.
