摘 要:目的研究不同镇痛药物对坐骨神经慢性压迫性损伤(chronic constriction injury,CCI)模型小鼠的镇痛作用,为神经病理性痛的临床治疗提供理论依据。方法采用8-0铬肠线限制性环扎小鼠右侧坐骨神经建立CCI神经病理性痛模型,用热板实验分别测定:不同剂量吗啡(10,5.0和2.5mg·kg-1,ip),曲马多(40,20和10mg·kg-1,ip),布洛芬(400,200和100mg·kg-1,po)和阿司匹林(400,200和100mg·kg-1,po)给药前后模型小鼠患侧脚掌的缩足潜伏期。结果腹腔注射10mg·kg-1和5.0mg·kg-1吗啡可以显著提高CCI模型小鼠患侧脚掌的缩足潜伏期,产生明显的镇痛作用,其作用维持时间分别为45和15min。腹腔注射40和20mg·kg-1曲马多也可产生明显的镇痛作用,但镇痛作用维持时间相对较短,分别为30和15min。相反,口服不同剂量布洛芬和阿司匹林对模型小鼠均未产生明显的镇痛作用。结论不同镇痛药物对CCI神经病理性痛模型小鼠的镇痛效果不同。Ohjective To evaluate the effects of different analgesics on mice with chronic constriction injury (CCI) of the sciatic nerve, and to provide a theoretical basis for the clinical therapy of neuropathic pain. Methods After the establishment of CCI neuro- pathic pain mouse by restrictive ligation of right sciatic nerve with 8-0 chronic catgut, the ipsilateral paw withdrawal latency of CCI mice before and after administration of different doses of morphine (10,5, 2.5 mg·kg^-1 ,ip), tramadol (40,20,10 mg·kg^-1 , ip) ,ibuprofen (400,200,100 mg·kg^-1 , po) and aspirin (400,200,100 mg·kg^-1 , po) were measured by the hot plate test. Results Intraperitoneal injection of 10 and 5 mg·kg^-1 morphine significantly increased the paw withdrawal latency of CCI mice, and the analgesic effect lasted 45 and 15 min,respectively. Intraperitoneal injection of 40 and 20 mg·kg^-1 tramadol also produced significant analgesic effect,but the effect maintained a relatively short times,i, e. 30 and 15 min, respectively. In contrast,oral administration of different doses of ibuprofen and aspirin did not show any analgesic effect on CCI mice. Conclusion Different analgesics showed different analgesic effects on CCI neuroDathic pain mice.
