摘  要:为探究石河子地区侵染南瓜和加工番茄的CMV卫星RNA及基因组RNA3的分子变异及株系分化,从石河子蔬菜研究所采集南瓜和加工番茄样品并提取dsRNA,利用RT-PCR方法克隆5个CMV卫星RNA序列和2个CMV RNA3序列,南瓜样品卫星RNA及RNA3分别命名为N-sate和N-R322,片段大小分别为335nt和2 214nt;加工番茄样品卫星RNA及RNA3分别命名为To-9-sate、S18-1、S9、S18-2和To-9-R322,片段大小分别为381nt、396nt、334nt、334nt和2 216nt。卫星RNA序列分析结果显示,卫星RNA的序列差异与地域有一定的相关性,与寄主类别无关。同时这5个卫星RNA序列都不含有CMVYSAT所具有的黄化区域。N-R322和To-9-R322序列分析结果显示,二者均属于CMV亚组ⅠB,且CMV分离物有一定的地域相关性,没有明显寄主相关性。In order to investigate the mutation and strain diversity of cucumber mosaic virus (CMV) infected the processing tomato and pumpkin in Shihezi of Xinjiang, five satellite RNA sequences and two RNA3 gene sequences were obtained by RT-PCR from pumpkin N-dsRNA and four processing tomato dsRNAs in vegetable research institute in Shihezi. The sequences obtained from pumpkin were named as N-sate and N-R322, with a length of 335 nt and 2 214 nt, respectively. The sequences from tomato were named as To-9-sate, S18-1, S9, S18-2 and To-9-R322, and the length was 381nt, 396 nt, 334 nt, 334 nt and 2 216 nt, respectively. Sequence analysis of satellite RNAs showed that the sequence had a certain correlation with regions, but not with the host, and also find that the five satellite RNA sequences do not contain yellow region of CMVYSAT. At the same time, the sequence analysis of N-R322 and To-9-R322 indicated that they belonged to CMV subgroup IB and the isolates of CMV had a certain correlation with regions, and they had no obvious correlation with the host.
