急性微循环障碍小鼠体内IL-17的表达和丹皮酚的作用

摘  要:目的研究急性微循环障碍小鼠体内IL-17的表达和分泌情况,以及丹皮酚的调控作用。方法用肾上腺素和冰水复制小鼠的急性微循环障碍模型,采用Real-time RT-PCR方法,检测组织中IL-17mRNA的表达和丹皮酚对其的抑制作用;应用酶联免疫吸附实验(ELISA)检测血清中IL-17的分泌和丹皮酚对其的抑制作用。结果肾上腺素和冰水浴所致急性微循环障碍,小鼠组织中IL-17mRNA的表达明显增加,血清中细胞因子IL-17的分泌明显增加,而给予丹皮酚治疗能明显抑制细胞因子IL-17mRNA的表达和血清中IL-17的分泌。结论急性微循环障碍小鼠体内IL-17表达和分泌明显增加,而丹皮酚可通过抑制IL-17表达和分泌来保护血管内皮损伤。Objective To study the expression of IL-17 in mice with acute microcirculation disorder, and analyze the regulatory role of paeonol on the expression of IL-17 in vivo. Methods The acute microcirculation disorder murine model was established by injection with adrenaline, irritated by cold- water. The mRNA expression of IL-17 in tissues was detected by real-time RT-PCR; the secretion of IL- 17 in the serum and the inhibition effect of paeonol on it were analyzed with ELISA. Results There was up- regulated mRNA expression of IL-17 in the involved tissues of the model group, and the concentration of IL-17 in serum was also increased in the model group. Pretreatment with paeonol could significantly suppress the expression of the cytokine, IL-17 mRNA, and the secretion of IL-17 in serum. Conclusion IL-17 expression was increased in the routine model with acute microcirculation disorder, and paeonol could be a promising drug to protect vascular endothelial cells against injuries through inhibition of IL-17 expression and secretion.
