单核细胞趋化蛋白-1及其受体与子痫前期发病关系

摘　　要：目的探讨单核细胞趋化蛋白-1(MCP-1)及其受体(CCR2)与子痫前期发病的关系。方法选择在我院住院分娩的子痫前期孕妇60例,其中重度子痫前期30例,轻度子痫前期30例,以同期分娩的正常晚期妊娠孕妇30例为对照组。采用酶联免疫吸附测定(ELISA)法检测两组孕妇血清MCP-1浓度,采用实时PCR技术检测两组孕妇胎盘组织中MCP-1及CCR2mRNA表达水平。结果子痫前期组孕妇血清MCP-1浓度显著高于对照组(t=3.4,P<0.05),轻度子痫前期组与重度子痫前期组比较,差异无统计学意义(P>0.05)。子痫前期组孕妇胎盘组织中CCR2mRNA的表达显著高于对照组(t=6.1,P<0.05),轻度子痫前期组与重度子痫前期组比较,差异无统计学意义(P>0.05)。子痫前期组孕妇胎盘组织中MCP-1mRNA的表达与对照组比较,差异无统计学意义(P>0.05)。结论孕妇血清中MCP-1及胎盘组织CCR2mRNA的表达与子痫前期发病密切相关,但与疾病严重程度无明显关系。Objective To study the association of monocyte chemotactic protein-1 （MCP-1） and its receptor （CCR2） with preeclampsia. Methods Sixty women with preeclampsia （30 severe and 30 mild） who gave birth in our hospital were selected, and 30 late pregnant women who gave birth in the same time period were served as controls. ELISA was used to measure the levels of serum MCP-1. The expressions of MCP-1 massager RNA（mRNA） and CCR2 mRNA in placenta tissue were determined by RT- PCR. Results In preeclampsia group, the serum concentration of MCP-1 was significantly higher than the control （t =3.4,P〈 0.05）, No significant difference was found in the expression of MCP-1 between severe and mild preeclampsia group （P 〈0.05）. CCR2 mRNA expression in placenta in preeclampsia group was significantly higher than the control （t= 6.1,P〈0.05）, bur that between severe and mild preeclampsia group was not statistically significant （P〉0.05）. No significant difference was found in the expressions of MCP 1 mRNA in placenta between preeclampsia and control group （P〈0.05）. Conclusion The expression levels of MCP-1 in serum and CCR2 mRNA in placenta were closely related to the occurrence of preeclampsia, but not with the severity of the illness.
