摘  要:目的研究明日叶查尔酮(AC)对2型糖尿病大鼠血管内皮细胞损伤的保护作用。方法将用高脂饲料喂养加链脲佐菌素腹腔注射诱发的2型糖尿病动物模型,随机分为糖尿病对照组和高、中、低剂量AC组,每组10只。糖尿病对照组喂饲高脂饲料并经口灌胃蒸馏水,高、中、低剂量AC组分别经口灌胃30、10和5mg/kg AC,正常对照组给予普通饲料,连续4周后测定各组血清内皮素-1、钙黏蛋白、丙二醛、空腹血糖和血清胰岛素等水平的变化。结果与糖尿病对照组比较,高剂量AC组内皮素-1和钙黏蛋白的含量降低,空腹血糖和血清胰岛素水平降低,差异均有显著性(F=4.58~48.29,q=3.45~14.94,P<0.05)。结论 AC可降低2型糖尿病大鼠血清内皮素-1和钙黏蛋白的水平,对血管内皮细胞的损伤有保护作用。Objective To study the protection of ashitaba chalcone (AC) on vascular endothelial cell injury in rats with type 2 diabetes mellitus (T2DM). Methods The rat models of T2DM-ereated by high-fat feeding plus intraperitoneal streptozo- tocin-were randomized to four groups as: control, high-dose-, middle-dose-and low dose-AC, with 10 rats in each group. The rats in diabetic-control group were fed with high-fat diet and intragastric administration of distilled water, those in high-dose-, middle- dose-, and high-dose-AC groups were respectively offered AC of 30, 10 and 5 mg/kg, intragastrically, and fed with common diet for four weeks. Serum endothelin-1 (ET-1), VE-ca, fasting serum glucose and serum insulin were then measured. Results Compared with the control, the levels of blood glucose, serum insulin, serum ET-1 and VE-ca in rats of high-dose-AC group decreased (F=4.58-48.29,q=3.45-14.94,P〈0.05). Conclusion AC may reduce the levels of serum ET-1 and VE ca and protect the endothelial cells of rats with type 2 diabetes.
