摘  要:目的探讨爆炸伤兔脑组织水通道蛋白-4(AQP-4)的表达变化及意义。方法将80只新西兰大白兔随机分为外伤组(70只)及对照组(10只),外伤组制备兔脑爆炸伤模型,对照组不进行任何干预。采用免疫组化法检测AQP-4蛋白水平,RT-PCR法检测AQP-4 mRNA水平。结果与对照组比较,外伤组AQP-4蛋白及AQP-4 mRNA水平随伤后时间延长而升高,伤后3 d达最高值,7 d后开始下降(P均<0.05)。结论爆炸伤后兔脑组织AQP-4表达升高,此可能与爆炸伤引起的继发性脑水肿密切相关。Objective To investigate the expression of aquaporin-4 (AQP-4) in the rabbit brain tissue with blast inju- ry and its significance. Method Eighty New Zealand white rabbits were randomly divided into the trauma group (n = 70) and the control group (n = 10). The rabbit brain blast injury model was prepared in the trauma group, and the control group without any intervention. The AQP-4 protein level was detected by the immunobistochemistry assay, and the AQP-4 mRNA level was detected by RT-PCR assay. Results Compared with the control group, the AQP-4 protein and AQP-4 mRNA levels in the trauma group were increased with the extension of time after injury, reached the highest value 3 d after injury, and 7 days later it began to decrease ( all P 〈 0.05 ). Conclusion The AQP-4 expression in rabbit brain after blast injury was increased, which may be closely related to the secondary brain edema.
