摘  要:目的:探讨白藜芦醇(resveratrol,RES)对缺氧(hypoxia)诱导神经元凋亡的影响。方法:原代培养小鼠皮层神经元,置于2%O2,5%CO2和93%N2,37℃培养箱中培养12 h建立缺氧模型,采用DCFH-DA和显微荧光成像系统检测神经元活性氧(Reactive oxygen species,ROS)含量;碘化丙啶(Propidium Iodide,PI)和Hoechst 33342染色检测神经元凋亡。结果:RES预处理60 min后再缺氧12 h,ROS含量和神经元凋亡率均显著低于缺氧组(P<0.01,N=3),且使用1 mM活性氧清除剂(N-acetyl-L-cysteine,NAC)也降低缺氧诱导的神经元凋亡率(P<0.01,N=6)。结论:RES抑制缺氧诱导皮层神经元凋亡,此作用可能与RES抑制缺氧所致神经元ROS增多有关。Objective:To explore the effects of resveratrol(RES) on neuronal apoptosis induced Hypoxia in cortical neurons.Methods: The present study established an hypoxic model in primary cultured cortical neurons of mice.In the hypoxia model,the level of ROS was determined using DCFH-DA and fluorescence microscopic imaging systems and apoptosis was analyzed by staining of propidium iodide(PI) and Hoechst 33342.Results: After pretreated with 25 μM RES for 60 min before hypoxia,the level of ROS and neuronal apoptosis were significantly lower than those in hypoxia cells.And pretreatment with 1 mM NAC(a ROS scavenger) can also reduce neuronal apoptosis percentage induced by hypoxia in neurons.Conclusion: The results indicated that RES could inhibit the hypoxia induced apoptosis in cortical neurons via reducing the level of ROS.
