摘 要:目的:研究黄连-石菖蒲抗糖尿病认知功能障碍的配伍规律,寻求最佳配伍比例.方法:分别建立四氧嘧啶致小鼠糖尿病模型和D-半乳糖致小鼠认知功能障碍模型(避暗法),分别观察黄连-石菖蒲不同配比改善糖代谢及认知功能的影响.结果:在降糖实验中,黄连石菖蒲药对5/0、4/1、3/2、2/3配比组能明显降低糖尿病小鼠FBG和GHb的含量,显示出较好的降糖活性;同时,在改善认知功能障碍的避暗实验中,黄连石菖蒲药对3/2、2/3、1/4、0/5配比组能明显延长小鼠的潜伏期,减少进入暗室的错误次数,显示出较好的改善认知功能,增强学习记忆能力的作用,尤以黄连-石菖蒲2:3配伍综合疗效更优.结论:黄连-石菖蒲药对具有明显的改善糖代谢和认知功能的作用,该作用是黄连-石菖蒲抗糖尿病认知功能障碍的药理学基础.Objective: To investigate the compatible regularity of drug pair of Rhizoma coptidis and Acorus gramineus on the treatment of diabetic cognitive dysfunction, and to find out the best compatibility proportion. Methods: Establishing alloxan induced diabetic mice model and D-galactose induced cognitive dysfunction model, and then observing the effects of different ratios of Rhizoma coptidis-Acorus gramineus on glucose metabolism and cognitive function, respectively. Results: Drug pair of Rhizoma coptidis and Acorus gramineus in the proportions of 5:0, 4:1, 3:2, 2:3 could significanly reduce the levels of fasting blood glucose (FBG) and Glycosylated hemoglobin (GHb) of diabetic mice. Meanwhile, drug pair of Rhizoma coptidis and Acorus gramineus also could obviously enhance the latency period and decrease the errors in entering the dark room in D-galact0se induced cognitive impairment mice. According to the analysis of comprehensive therapeutic effects on diabetes and cognitive dysfunction, the optimal proportion of Rhizoma coptidis and Acorus gramineus is 2:3. Conclusion: Drug pair of Rhizoma eoptidis and Acorus gramineus possess visible improving effects on glucose metabolism and cognitive function.
