摘 要:目的:研究新型氟喹诺酮类药物盐酸安妥沙星(antofloxacin hydrochloride,ANTO)对金黄色葡萄球菌和大肠埃希菌的持续效应:抗生素后效应(PAEs)、抗生素后亚抑菌浓度效应(PASMEs)和抗生素亚抑菌浓度效应(SMEs)。方法:采用宏量肉汤稀释法测定最小抑菌浓度(minimal inhibitory concentrations,MIC)。35℃环境中细菌暴露于抗菌药物2h后用稀释法去除抗菌药物,以活菌计数法计数作细菌生长曲线,计算PAEs、PASMEs和SMEs。结果:盐酸安妥沙星对金黄色葡萄球菌和大肠埃希菌均呈现明显的PAEs、PASMEs和SMEs,且PASMEs和SMEs均呈现明显的浓度依赖特点。结论:盐酸安妥沙星对金黄色葡萄球菌和大肠埃希菌均可产生明显的PAEs、PASMEs和SMEs,提示制定给药方案时可考虑适当延长给药时间间隔。Objective: To study the sustained effects of new fluoroquinolones antofloxacin hydrochloride against Staphylococcus aureus and Escherichia coli,which consist of post antibiotic effects (PAEs) 、post antibiotic subminimum inhibitory concentration effects (PASMEs) and Sub-inhibitory Effects(SMEs) . Methods: Minimal inhibitory concentrations(MIC) were determined by broth macro-dilution method. The antimicrobial agents were eliminated by dilution method after the bacteria were exposed to antimicrobials for 2h. Growth curves were followed by viable counts,and then PAEs、PASMEs and SMEs were calculated. Results: Antofloxacin hydrochloride had significant PAEs、PASMEs and SMEs against Staphylococcus aureus and Escherichia coli,and PASMEs and SMEs were distinct concentrationdependant. Conclusion: As the PAEs、PASMEs and SMEs induced by antofloxacin hydrochloride were obvious. It suggests that when preparing a prescription,we could prolong the medicine-administered interval time properly.
