摘 要:目的构建TatABC基因缺失株,并探讨其与肠侵袭性大肠杆菌(EIEC)生理功能的关系。方法利用一步法基因缺失方法,敲除EIEC菌株的TatABC基因,探索该基因对EIEC形态、生长、生化及跨膜转运等生理功能的影响。结果TatABC基因缺失株的EIEC在镜下呈链状排列,在LB培养基上生长显著变慢;生化反应无明显变化;在厌氧条件下,由于不能转运TMAO还原酶,失去了在M9培养基上下生长的能力。结论 EIEC的Tat蛋白运输系统与其生理功能密切相关。Objective To construct the deletion mutant for TatABC gene of EIEC and explore the tat system effect on its physiological function. Methods The TatABC gene was deleted by using single step PCR method, and its effect on the EIEC morphology, growth rate, biochemical reaction and membrane transport were observed. Results Significant differences existed between the TatABC mutant strain and wild type strain . First, the TatABC mutantstrain arranged in chains and grew slowly in LB medium. Second, no significant changes in biochemical reactions ap- peared between the wild type and mutant strain. Third, because of the failure to translocate TMAO reductase, the mutant strain could not grow anaerobically with M9 culture. Conclusion These results establish that TatABC gene has a profound effect on EIEC physiological function.
