摘  要:应用地高辛标记技术,对通过睾丸注射法制备的转基因猪进行荧光原位杂交,确定外源pGH(猪生长激素)基因在染色体上的整合位点及数目。选用睾丸注射法制备的140日龄转基因长白猪6头与同期同龄非转基因猪2头进行血细胞培养及荧光原位杂交。结果显示,外源基因已整合于染色体上,但在染色体上的整合位点具有随机性,而且不同的转基因猪阳性个体外源基因在染色体上的整合位点有差异,但集中分布于1、6、8、13号染色体上;非转基因猪染色体上无杂交信号;个体杂交信号数越多,其生长速度越快。Digoxin labeling technology was used to transgenic pigs, which was processed by testicu- lar injection method,and then used fluorescence in situ hybridization (FISH) on the transgenic pigs,which was in order to confirm the numbers and integration sites of foreign gene pGH( por- cine growth hormone) on chromosomes. Cultivation of blood cell and FISH were proceeded in 6 transgenic Landrace pigs and 2 non-transgenic Landrace pigs, which were 140 days obtained by testicular injection method. The foreign gene had been integrated on chromosomes,not only the randomness of location was found on chromosomes,but also that different positive transgenic in- dividuals had different location was proved. Several chromosomes like number 1,6,8 and 13 were intensivly found to have the location lbut the chromosomes of non-transgenic Landrace pigs do not have hybridization signal, and the greater hybridization signals between individuals were, the more obvious diffence of the growth rate was. The above results will offer a basic foundation of the cor- relation between foreign gene and other relevant functional genes on the same chromosome.
